【摘要】:微流控芯片或芯片实验室是系统生物学研究的重要技术平台之一。本论文以微流控芯片为技术依托,从具有临床意义的疾病诊断出发,对微流控芯片在基因分析和细胞凋亡等领域的应用进行了一系列具有一定覆盖面的研究,力图发展具有潜在生物医学应用价值的相关技术。论文共分六章。  第一章阐述了微流控芯片在基因分析和细胞凋亡领域研究的进展。  第二~五章集中探讨了利用实验室自行设计、研制的微流控芯片系统,开展以基因多态性分析、外源性基因检测和基因表达为主要途径的基因分析的可行性。主要包括:1)大连地区123 例高血压患者血管紧张素原基因启动子区A-6G基因多态性分析;2)人参微卫星位点短串联重复序列多态性分析,以及在此基础上的东方参和西洋参鉴别;3) 临床常见病原体结核杆菌、丙型肝炎病毒基因PCR 产物和人类白细胞相关抗原(HLA-B27)基因的快速检测;和4)细胞RNA完整性评价及不同组织岩藻糖基转移酶基因表达分析。 鉴于细胞凋亡这一生物学过程在疾病诊断和治疗上的重要意义,论文第六章以氧化砷诱导急性早幼粒白血病细胞凋亡为模型,以细胞氧化还原状态变化和DNA 片段化等凋亡事件为主要研究内容,实现了对凋亡白血病细胞内活性氧和还原型谷胱甘肽的同时测定以及DNA 片段化分析,显示了微流控芯片平台用于细胞凋亡研究的良好前景。 
[Abstract]:Microfluidic chip or chip laboratory is one of the important technology platform for systems biology research. In this paper, the microfluidic chip technology, starting from the diagnosis of clinical significance, the application of microfluidic chip in gene analysis in the field have carried out a series of certain coverage of the study, in order to develop the related techniques with potential biomedical application value. This dissertation consists of six chapters.
The first chapter describes the progress of microfluidic chips in the field of gene analysis and cell apoptosis.
Second to the five chapter discusses the use of self-designed laboratory, microfluidic chip system development, to carry out genetic polymorphism analysis. The expression of exogenous gene detection and gene feasibility analysis as the main pathway. Mainly includes: 1) 123 cases of hypertension patients in Dalian region of angiotensinogen gene analysis of the promoter region of A-6G gene polymorphism; 2) repeat polymorphism analysis of microsatellite loci in short series, and on the basis of the Oriental ginseng and American ginseng identification; 3) fromtuberculosis, hepatitis C virus PCR gene product and human leukocyte antigen (HLA-B27) for rapid detection of gene expression analysis; and 4) enzyme gene transfer of RNA cells in different tissues and evaluate the integrity of fucosylation.
In view of the important significance of cell apoptosis in the biological process in the diagnosis and treatment of diseases, the sixth chapter of the thesis to arsenic induced apoptosis of acute promyelocytic leukemia cells as a model, the cellular redox state changes and DNA fragmentation of apoptotic events as the main research contents, the analysis on the determination of active oxygen in the apoptosis of leukemia cells at the same time, glutathione and DNA fragmentation, good prospects for apoptosis research of microfluidic chip platform for display.
