【摘要】: 本研究建立了烟曲霉感染的动物模型,筛选出强、弱毒株,利用比较蛋白质组学技术,分析了两者的蛋白质表达谱,发现强、弱毒株之间蛋白质组成存在差别,经软件分析匹配选取39个经质谱检测,鉴定出6个蛋白质。其中,强毒株中有2个表达上调的蛋白质(Cat1P,AspF3)认为与致病性相关;相应基因分析结果表明蛋白质的这种表达差异主要是在转录水平发生的。在弱毒株中表达上调的蛋白质有4个(actin,NADP linked glutamate dehydrogenase , guanine nucleotide-binding beta subunit liked protein,orotidine 5`-phosphate decarboxylase)分别涉及细胞活动、代谢合成及信号传导途径的某个环节。本研究还进行了烟曲霉与念珠菌蛋白质组的二维凝胶电泳图谱比较,显示一些主要高丰度表达蛋白质不在同一区域内,认为它们的蛋白质组存在明显差异。本实验为烟曲霉致病机制的研究、抗真菌药物及疫苗的研发提供了重要的理论基础和实验依据。
[Abstract]:In this study, the animal model of Aspergillus fumigatus infection was established, and the strong and weak strains were screened. The protein expression profiles of the two strains were analyzed by comparative proteomics technique, and the difference of protein composition between the strong and weak strains was found. By software analysis, 39 proteins were identified by mass spectrometry. Among them, 2 of the virulent strains expressed up-regulated protein Cat1PAspF3, which were thought to be related to pathogenicity. The results of gene analysis showed that the difference of protein expression was mainly at the transcriptional level, and four proteins were up-regulated in attenuated strains. NADP linked glutamate dehydrogenase. Guanine nucleotide-binding beta subunit liked protein. Orotidine 5 '-phosphate decarboxylase is involved in cell activity. This study also compared the proteome of Aspergillus fumigatus and candida by two-dimensional gel electrophoresis, which showed that some of the major high abundance proteins were not in the same region. This study provides an important theoretical basis and experimental basis for the study of pathogenic mechanism of Aspergillus fumigatus and the research and development of antifungal drugs and vaccines.
