人胎儿卵巢体外培养的实验研究

【摘要】:目的:探讨人胎儿卵巢在体外培养及冷冻保存复苏后不同条件下卵泡发育并分泌雌激素情况,探讨体外培养条件下卵泡刺激素(follicle stimulating hormone,FSH)、N-乙酰-L-半胱氨酸(N-acetyl-L-cysteine,NAC)对人胎儿卵巢发育的影响;了解人胎儿卵巢组织的最佳体外培养条件,为后续研究提供必要的实验依据。 方法:选取20~28周的胎儿卵巢30例采用器官组织培养的方法进行体外培养,按照培养液不同成分分为O组(O液:生理盐水)、A组(A液:生理盐水加20%新生牛血清)、B组(B液:基本培养液)、C组(C液:含0.1mmol/l NAC的基本培养液)、D组(D液:含0.5u/ml FSH的基本培养液)、E组(E液:含0.1mmol/l NAC、0.5u/ml FSH的基本培养液)每3天更换培养液并保存培养液以液相平衡竞争放射免疫分析法(RIA)进行雌二醇(estradiol,E_2)测定,并留取培养后人胎儿卵巢组织块进行组织学观察。 结果:1.相同培养时间下,B组卵巢雌二醇分泌量高于O组、A组,二者相比差异显著(P0.05);O组与A组,培养后第4~6天、7~9天卵巢雌二醇分泌量低于第1~3天卵巢雌二醇分泌量,二者差异显著(P0.05);B组培养第4~6天、7~9天卵巢雌二醇分泌量高于第1~3天分泌量,二者相比差异显著(P0.05)。2.相同培养时间下,新鲜组与冻融组之间,卵巢雌二醇分泌量无显著性差异(P0.05),相同培养时间下,新鲜组与冻融组之间,卵巢各期卵泡构成比无显著性差异(P0.05);而新鲜组与冻融组内培养第4~6天、7~9天卵巢雌二醇分泌量高于第1~3天卵巢雌二醇分泌量,二者差异性显著(P0.05)。新鲜组与冻融组内,培养第3天、6天、9天后卵巢中较为成熟的卵泡比例明显高于未培养组(P0.05)。3.相同培养时间下,C组、D组、E组卵巢雌二醇分泌量高于B组,二者差异性显著(P0.05)。C组、D组、E组内,培养第4~6天、7~9天卵巢雌二醇分泌量高于第1~3天卵巢雌二醇分泌量,二者差异性显
[Abstract]:Objective: To explore the human fetal ovary in vitro and frozen thawed under different conditions of follicular development and the secretion of estrogen, to investigate the in vitro follicle stimulating hormone (follicle stimulating, hormone, FSH), N- acetyl -L- cysteine (N-acetyl-L-cysteine, NAC) on human fetal ovarian development; to understand the optimal culture conditions in vitro of human fetus ovarian tissue, provide the necessary experimental basis for further study.
Methods: the fetal ovary from 20 to 28 weeks, 30 cases with organ tissue culture method in vitro and in accordance with the different medium components were divided into O group (O solution: physiological saline), A group (A solution: normal saline and 20% newborn calf serum), B group (B solution: basic medium), group C (basic medium C liquid containing 0.1mmol/l NAC), D group (basic medium D liquid containing 0.5u/ml FSH), E group (basic medium E solution containing 0.1mmol/l NAC, 0.5u/ml: FSH) replaced the culture medium every 3 days and the preservation of culture liquid in liquid phase equilibrium competition radiation immunoassay (RIA) of estradiol (estradiol, E_2) were determined, and take the cultivation of human fetal ovarian tissue blocks for histological observation.
