【摘要】:[目的] 对日本血吸虫云南品系未成熟卵可溶性抗原(soluble immature eggs antigen,SIEA)免疫鼠体内日本血吸虫成虫的睾丸及卵黄腺组织进行超微结构观察,探讨SIEA抗日本血吸虫生殖作用机制;观察日本血吸虫云南品系未成熟卵可溶性抗原(SIEA)的抗病免疫作用;观察小鼠经云南品系SIEA免疫后用皖鄂品系血吸虫尾蚴攻击感染,是否也能诱导产生抗病免疫效果。 [方法] 制备日本血吸虫云南品系未成熟卵可溶性抗原(SIEA)。取适量SIEA加等量福氏佐剂,背部皮下多点注射免疫ICR小鼠后分2组,分别用大理钉螺和华东钉螺逸出的血吸虫尾蚴(30±2条)攻击感染。于感染后46天剖杀小鼠,观察减虫率、粪卵减少率、雌虫子宫内虫卵数、肝表面虫卵结节数、肝脏各期虫卵数及各期虫卵构成比;取小鼠肝脏按常规制作病理切片,观察肝组织内肉芽肿病变情况并分析肉芽肿平均直径;取合抱成虫,分离出雌虫卵黄腺和雄虫睾丸组织进行超微结构观察。 [结果] SIEA免疫后减虫效果不明显,大理组与华东组粪卵减少率分别为33.25%、34.45%(P0.05),雌虫子宫内虫卵数减少率分别为34.75%、23.69%(P0.05),肝表面虫卵结节密度分别下降47.76%、29.03%(P0.05),肝虫卵肉芽肿大小比对照组分别下降了49.06%、30.69%(P0.05);肝组织内虫卵总数下降,大理组与华东组每雌肝卵减少率分别为
[Abstract]:[Objective] to Yunnan strains of Schistosoma japonicum soluble immature egg antigen (soluble immature eggs antigen, SIEA) on the ultrastructure of mice immunized Schistosoma japonicum adult testis and yolk gland tissue, SIEA of Schistosoma japonicum reproductive mechanism; observe the Yunnan strain of Schistosoma japonicum soluble immature egg antigen (SIEA) immune disease effect; mice immunized with SIEA Yunnan strains with Anhui and Hubei strains of schistosoma cercariae infection, can induce the immune effect of resistance.
[method] the preparation of Yunnan strains of Schistosoma japonicum soluble immature egg antigen (SIEA). Apply SIEA with equal volume of Freund's adjuvant. Subcutaneous multi-point injection of immune ICR mice were divided into 2 groups, respectively in Dali and East China from Oncomelania snails schistosoma cercariae (30 + 2) infection after infection 46 attack. The day the mice were sacrificed and the observation of the worm reduction rate, fecal egg reduction rate, the number of eggs within the female uterus, liver surface egg nodule number, the number of eggs in liver ratio and different stages of egg; mice liver by routine pathological sections were made, were observed in liver tissue and granuloma granuloma analysis of average diameter; take the paring adult isolated, ultrastructure of testicular tissue of the vitelline gland of female and male.
[results] SIEA after immunization with the worm reduction effect is not obvious, Dali group and East Group fecal egg reduction rates were 33.25%, 34.45% (P0.05), the number of eggs within the female uterus reduction rate were 34.75%, 23.69% (P0.05), the surface of the liver egg nodule density decreased by 47.76%, 29.03% (P0.05), hepatic granuloma size compared with the control group decreased by 49.06%, 30.69% (P0.05); liver eggs in the decline in the total number of Dali group and East Group, each female liver egg reduction rates were
