【摘要】： 心肌肌钙蛋白是心肌运动的调节蛋白,心肌梗死等损伤性疾病发生时,其亚单位肌钙蛋白T和肌钙蛋白I释放入血,是诊断心肌损伤的特异性标志物。本研究通过组织匀浆、蛋白层析技术实现对牛心肌肌钙蛋白T的提取和纯化,通过SDS-PAGE和ELISA方法鉴定获得蛋白的性质。获取的蛋白免疫动物后,采用经典的细胞融合技术,制备杂交瘤细胞,通过ELISA方法鉴定培养细胞上清,经杂交瘤细胞克隆,分选,扩增并稳定冻存,获得5株阳性细胞株,为心肌肌钙蛋白检测试剂的国产化奠定实验基础。
[Abstract]:Cardiac troponin is the regulating protein of myocardial movement. When myocardial infarction and other injury diseases occur, its subunit troponin T and troponin I release into the blood. In this study, bovine cardiac troponin T was extracted and purified by tissue homogenate and protein chromatography. The obtained protein was immunized by SDS-PAGE and ELISA. The hybridoma cells were prepared by classical cell fusion technique. The supernatants of cultured cells were identified by ELISA method, cloned, sorted, amplified and frozen stably by hybridoma cells, and 5 positive cell lines were obtained, which laid the experimental foundation for the localization of cardiac troponin detection reagent.
