【摘要】: 目的:1.本研究拟通过检测吗啡戒断对小鼠自发活动的影响以期验证旷场实验作为研究吗啡戒断伴焦虑样行为动物模型的有效性。2.检测吗啡戒断对小鼠场景性恐惧条件化的影响。 方法:以剂量递增法连续皮下注射(sc)盐酸吗啡7d建立吗啡依赖小鼠模型。动物随机分为吗啡戒断1d组及其生理盐水对照组、吗啡戒断7d组及其生理盐水对照组、吗啡戒断21d组及其生理盐水对照组。各组动物在相应时间点采用旷场实验对自发活动(穿梭格数、直立次数、理毛时间、进入中央次数、中央区活动时间及外周区活动时间等指标)进行检测;随后进行场景性恐惧条件化训练,并与24h后进行条件化恐惧反应(僵直)检测。 结果:吗啡戒断1d组小鼠的穿梭格数、直立次数、进入中央区次数及中央区活动时间对比对照组均显著降低(p0.001,p0.001,p0.001,p0.001),而理毛时间和外周区活动时间与其对照组相比显著增加(p0.05,p0.01)。吗啡戒断7d组小鼠的穿梭格数、进入中央区次数及中央区活动时间与对照组比较显著降低(p0.05,p0.05,p0.05),同时外周区活动时间显著增多(p0.05),但是直立次数及理毛时间与其对照组比较没有显著性差异(p㧐0.05, p㧐0.05)。吗啡戒断21d组小鼠各项检测指标与其对照组比较差异均无显著性(p㧐0.05, p㧐0.05, p㧐0.05, p㧐0.05, p㧐0.05, p㧐0.05)。另外,与生理盐水对照组比较,吗啡戒断1d组小鼠的僵直百分比显著下降(p0.05),而吗啡戒断7d组和吗啡戒断21d组小鼠的僵直百分比与各自对照组比较无显著差异(p㧐0.05, p㧐0.05)。 结论:1.吗啡依赖小鼠在戒断早期其主要自发活动指标均发生显著性改变;随着戒断期延续,其自发活动逐渐恢复正常。提示旷场实验可以作为研究吗啡戒断早期伴焦虑样行为的有效动物模型。2.吗啡戒断可时间依赖性损伤小鼠场景性恐惧条件化。
[Abstract]:Objective: 1., the purpose of this study is to detect the effect of morphine withdrawal on spontaneous activity in mice, so as to verify the validity of open field test as a model for morphine withdrawal and anxiety like behavior..2. was used to detect the effect of morphine withdrawal on the conditioning of scene fear in mice.
Methods: with the increasing dosage by subcutaneous injection of morphine hydrochloride (SC) 7d in morphine dependent mice. Animal model were divided into 1D group and morphine withdrawal saline control group, morphine withdrawal group 7d and normal saline control group, morphine withdrawal group 21d and saline control group. Each animal by open field experiment on spontaneous activity in the corresponding time points (shuttle cell number, vertical frequency, grooming time, enter the central frequency, central activity time and the peripheral zone time index) were detected; then the contextual fear conditioning training, and the conditioned fear response and 24h (stiff) detection.
Results: the number of shuttle, lattice of morphine withdrawal mice in 1D group rearing, in contrast the central frequency and central time control group decreased significantly (p0.001, p0.001, p0.001, p0.001), and the hair was significantly increased compared with time and peripheral activity time and the control group (P0.05, P0.01). The number of shuttle lattice morphine withdrawal of mice in the 7d group, entered the central frequency and central time compared with the control group decreased significantly (P0.05, P0.05, P0.05), and peripheral zone activities increased significantly (P0.05), but the rearing and grooming time compared with the control group had no significant difference (P? 0.05, p? 0.05) there were no significant differences. Compared with morphine withdrawal mice in group 21d of the indexes and the control group (P? 0.05, P? 0.05, P? 0.05, P? 0.05, P? 0.05, P? 0.05). In addition, compared with saline control group, the percentage of stiff morphine withdrawal mice in group 1D decreased significantly (P 0.05) there was no significant difference in the percentage of rats in the morphine withdrawal group 7d and the morphine abstinence 21d group (P? 0.05, P? 0.05).
Conclusion: in the early withdrawal of the main indicators of spontaneous activity changes of 1. morphine dependent mice; with the withdrawal period of continuation of the spontaneous activity gradually returned to normal. That open field test can be used as.2. effective animal model of morphine withdrawal early with anxiety like behavior of morphine withdrawal time dependent contextual fear conditioning injury in mice.
