【摘要】:目的:比较脐带血细胞组与成人外周血细胞组天然免疫功能及各组红细胞对白细胞天然免疫功能调控能力的差异。方法:通过肿瘤血细胞花环实验测定脐带血细胞与成人外周血细胞的天然免疫黏附肿瘤细胞能力变化的指标,以及通过流式细胞仪法测定脐带血细胞与成人血细胞分别在全血细胞添加肿瘤细胞、全血细胞添加生理盐水、全血细胞去除红细胞后添加肿瘤细胞、全血细胞去除红细胞后添加生理盐水四种情况下CD35、CXCR4的荧光强度。结果:脐带血红细胞肿瘤黏附率、淋巴细胞肿瘤黏附率、粒细胞肿瘤黏附率分别低于对应正常成人组,统计学有差异;同样条件下成人红细胞CD35荧光强度要低于脐血组,而成人淋巴细胞、粒细胞CD35、CXCR4荧光强度要高于脐血组;成人与脐血淋巴细胞、粒细胞在添加了红细胞后CD35、CXCR4荧光强度要高于未添加红细胞组。结论:脐带血细胞天然免疫功能较健康成人外周血细胞天然免疫功能低;成人外周血及脐带血细胞中红细胞对白细胞的天然免疫功能皆有增强作用,在添加了肿瘤细胞等抗原刺激物表现更明显,其中脐带血红细胞的调控能力较正常成人外周血红细胞的调控能力要弱。
[Abstract]:Objective: to compare the difference of innate immune function between cord blood cell group and adult peripheral blood cell group and the regulation ability of red blood cell to leukocyte innate immune function. The changes of the ability of cord blood cells and adult peripheral blood cells to adhere to tumor cells were measured by rosette assay. Umbilical cord blood cells and adult blood cells were added tumor cells to whole blood cells, normal saline to whole blood cells, and tumor cells were added to whole blood cells after removing red blood cells by flow cytometry. The fluorescence intensity of CD35 and CXCR4 in the whole blood cells after removing red blood cells and adding normal saline. Results: the adhesion rate of cord blood red blood cell tumor and lymphocyte tumor. The adhesion rate of granulocyte tumor was lower than that of the corresponding normal adult group (P < 0.05). Under the same condition, the fluorescence intensity of CD35 in adult erythrocytes was lower than that in cord blood group, while the fluorescence intensity of CD35 and CXCR4 in adult lymphocytes was higher than that in cord blood group. In adults with cord blood lymphocytes, granulocytes were added with red blood cells after CD35. Conclusion: the innate immune function of cord blood cells is lower than that of healthy adult peripheral blood cells. Red blood cells in adult peripheral blood and umbilical cord blood cells have enhanced the innate immune function of leukocytes, especially in the addition of tumor cells and other antigenic stimulators. The control ability of umbilical cord blood red blood cell is weaker than that of normal adult peripheral blood red blood cell.
