骨髓间充质干细胞定向诱导分化成肌细胞的对比实验研究

【摘要】:骨骼肌在严重损伤后,由于肌细胞不能再生,其修复方式主要通过瘢痕修复,愈合后收缩功能差,骨髓间充质干细胞(mesenchymal stem cells,MSCs)作为成体干细胞的一种,具备多项分化潜能,在肌组织创伤修复方面具有重要作用。如何将MSCs定向诱导分化为完美成熟的肌细胞是目前正在研究的问题。 我们在成功分离并培养MSCs后,分别通过5-氮杂胞苷及骨骼肌损伤动物模型的不同时期血清连续诱导两种方法对MSCs进行诱导,并将诱导后的MSCs局部注射入损伤的肌组织中,观察其在损伤重建中发挥的作用。结果表明两种方法均能将MSCs诱导成肌细胞,而血清诱导组肌细胞发育更成熟。认识到,5-Aza作为单一的化学信号与损伤动物血清的生理性调节信号相比较,在MSCs分化成骨骼肌细胞方面存在一定差距。MSCs最终要分化为成熟的目的细胞,其诱导方式是非常复杂的。同时证明了应用肌肉损伤动物不同时期血清连续诱导MSCs向肌细胞分化成为可能,提出MSCs诱导分化的一个新途径。
[Abstract]:After severe injury, skeletal muscle cells can not regenerate, its repair way is mainly through scar repair, the contraction function is poor after healing. As a kind of adult stem cells, bone marrow mesenchymal stem cells have multiple differentiation potential. It plays an important role in wound repair of muscle tissue. How to induce and differentiate MSCs into perfect mature myocytes is a problem that is currently being studied. After successfully isolating and culturing MSCs, MSCs was induced by 5-azacytidine and serum at different stages of skeletal muscle injury. The induced MSCs was injected into the injured muscle tissue to observe its role in the reconstruction of the injury. The results showed that both methods could induce MSCs into muscle cells. However, the development of muscle cells in serum-induced group was more mature. It was recognized that 5-Aza as a single chemical signal was compared with the physiological regulatory signal of injured animal serum. There is a certain gap in the differentiation of MSCs into skeletal muscle cells. It is proved that it is possible to continuously induce the differentiation of MSCs into myocytes by serum in different stages of muscle injury, and a new way of inducing differentiation by MSCs is put forward.
