【摘要】:天然自身抗体(natural autoantibody,NAA)可以定义为在没有任何抗原免疫的情况下,正常机体产生的针对一种或多种自身抗原的抗体。NAA是构成天然免疫的重要组成部分,可以维持自身稳定、防止自身免疫病发生,具有抗炎、抗肿瘤监视作用等。但由于相关研究进展及实验方法的限制,对NAA的产生机制、产生NAA的自身反应性B细胞的选择/耐受机理等方面尚知之甚少。 抗体转基因小鼠的应用,为深入研究NAA产生机制提供了理想的平台。目前已建立的多种自身抗体转基因小鼠,从不同侧面揭示了自身耐受的部分机制,初步探讨了NAA的产生机理。本研究组对NAA家族之一的天然抗角蛋白自身抗体(anti-keratin autoantibody,AK auto Ab)进行了多年深入、细致的研究,探讨了AK auto Ab的许多重要功能。在此基础上,应用未免疫小鼠B细胞与骨髓瘤细胞融合,我们成功筛选到了分泌抗角蛋白抗体的杂交瘤,由未免疫小鼠获得的AK
[Abstract]:Natural autobody NAA) can be defined as immune without any antigen. As an important part of innate immunity, NAA produced by normal body against one or more autoantigens can maintain self-stability, prevent autoimmune diseases, and have anti-inflammation. However, the mechanism of NAA production and the selection / tolerance mechanism of NAA autoreactive B cells are not well understood due to the progress of related research and the limitation of experimental methods. The application of antibody transgenic mice provides an ideal platform for further study on the mechanism of NAA production. A variety of autoantibody transgenic mice have been established at present. Some mechanisms of self tolerance were revealed from different aspects. The production mechanism of NAA was preliminarily discussed in this study. This study aimed at anti-keratin autoantibody, one of the natural anti-keratin autoantibodies of NAA family. AK auto Ab) has been studied deeply for many years, and many important functions of AK auto Ab have been discussed. Using the fusion of B cells and myeloma cells of unimmunized mice, we successfully selected hybridomas secreting anti-keratin antibodies, and obtained AK from unimmunized mice.
