【摘要】: 神经老化是生命活动的必然,探讨神经老化机制,寻找延缓神经老化进程的方法是神经科学领域里的一个重要内容。在脑衰老研究中,突触可塑性备受人们关注。δ-连环素(δ-catenin)与树突棘素(Spinophilin)是近年来发现的分布于神经元树突的蛋白,在突触塑形中扮演重要角色。δ-catenin是p120-catenin (p120ctn)蛋白家族成员,具有能和钙粘素(cadherins)近膜端结合的序列,δ-catenin是这一蛋白家族唯一特异性只表达于神经系统的蛋白,它能和突触后成分蛋白的PDZ结构域结合。δ-catenin定位于突触后区域,能调节突触的传入和塑形。Spinophilin因其特异性定位于树突棘头部,故得名树突棘素。它具有细胞连接处绞手架蛋白(scaffold protein)的特征,并且含有和PSD95/DLG/zo-1相同的序列,是蛋白磷酸酶-1(PP1)新的靶向亚单位,能将PP1定位于树突棘的底物(如受PP1调节,从而影响突触功能的神经递质受体)上,调节树突棘的形态,完成突触塑形。小脑是脑衰老机理研究的理想模型系统之一。 本实验在建立大鼠自然老化模型的基础上,运用免疫组织化学方法检测δ-catenin及spinophilin在不同年龄大鼠小脑的定位表达;运用图像分析系统和免疫印迹(Western-blot)技术检测δ-catenin及spinophilin在大鼠小脑的表达水平;运用RT-PCR技术检测δ-catenin,spinophilin mRNA在不同年龄组大鼠小脑表达的差异。 免疫组织化学检测显示δ-catenin,spinophilin主要表达在小脑分子层和颗粒层,在中、青年大鼠蒲肯野细胞有中等阳性表达,老年大鼠蒲肯野细胞表达较弱。Western blot检测显示δ-catenin、spinophilin在中年组大鼠小脑表达最高,在老年组大鼠小脑表达最低。RT-PCR检测显示δ-catenin、spinophilin mRNA表达水平的增龄性变化与其蛋白表达的增龄性变化一致。 本实验研究结果表明δ-catenin,spinophilin在大鼠小脑的表达存在增龄性变化。这一变化和不同年龄大鼠小脑突触的数量,结构和功能的变化有关。
[Abstract]:Nerve aging is a necessity of life activity. It is an important content in the field of neuroscience to explore the mechanism of nerve aging and to find ways to delay the process of nerve aging. Synaptic plasticity has attracted much attention. 未 -catenin (未 -catenin) and spinophilin (Spinophilin) are proteins found in recent years. 未 -catenin is a member of p120-catenin / p120ctnprotein family. 未 -catenin is the only protein expressed specifically in the nervous system with a sequence that binds to the proximal end of the cadherins. It can bind to the PDZ domain of the postsynaptic component protein. 未 -catenin is located in the postsynaptic region. It can regulate the afferent and shape of synapses. Spinophilin is located in the head of dendritic spine because of its specificity. It has the characteristics of scaffold protein (scaffold protein) and contains the same sequence as PSD95/DLG/zo-1. It is a new target subunit of protein phosphatase (PP-1), which can locate PP1 on the substrate of dendritic spine (such as neurotransmitter receptor, which is regulated by PP1, which affects synaptic function). The cerebellum is one of the ideal model systems for the study of brain aging mechanism. The expression of 未 -catenin and spinophilin in cerebellum of rats of different ages was detected by immunohistochemical method on the basis of establishing natural aging model of rats. The expression of 未 -catenin and spinophilin in cerebellum of rats was detected by image analysis system and Western-blot technique. The expression of 未 -cateninine spinophilin mRNA in the cerebellum of rats of different age groups was detected by RT-PCR technique. Immunohistochemical examination showed that 未 -cateninine spinophilin was mainly expressed in the molecular and granular layers of cerebellum, and the expression of 未 -cateninine spinophilin was moderately positive in the young rats' Purkinje cells. The expression of 未 -catenin spinophilin was the highest in the middle aged rats. The lowest expression of 未 -catenin was detected by RT-PCR in the cerebellum of the aged rats. The age change of spinophilin mRNA expression level was consistent with that of its protein expression. The results showed that the expression of 未 -cateninine spinophilin in the cerebellum of rats had age-related changes and the number of cerebellar synapses in rats of different ages. Changes in structure and function are relevant.
