miR-124a通过抑制iASPP基因表达促进神经突起生长

【摘要】:目的探索miR-124a的靶基因iASPP对神经发育的影响。方法利用在线预测软件TargetScan对miR-124a的靶基因进行预测,结合文献资料提到的与神经早期发育有关的基因进行筛选,我们选择iASPP作为miR-124a的候选靶基因。通过荧光素酶报告基因实验对其进行验证,然后在M17细胞中转染miR-124a过表达质粒,用Western blotting实验检测iASPP蛋白表达水平的变化。利用视黄酸诱导M17细胞作为神经元分化模型,通过过表达或者基因干扰的方法,初步探讨iASPP基因对神经发育的影响。结果 miR-124a促进神经突起发育,并且能够与iASPP基因的3'非翻译区(3'UTR)相互作用来抑制其蛋白质的表达;iASPP基因的过表达抑制神经突起的发育并且抑制miR-124a促进神经突起生长的作用。结论 miR-124a能够通过抑制iASPP基因的表达来促进神经突起生长。
[Abstract]:Objective to explore the effect of target gene iASPP of miR-124a on neural development. Methods the target gene of miR-124a was predicted by online prediction software TargetScan. We selected iASPP as the candidate target gene for miR-124a, verified it by luciferase reporter gene experiment, then transfected miR-124a overexpression plasmid in M17 cells. The expression of iASPP protein was detected by Western blotting assay. M17 cells were induced by retinoic acid as a neuronal differentiation model. The effect of iASPP gene on nerve development was studied. Results miR-124a promoted neurite development. And it can interact with the 3'untranslated region of iASPP gene to inhibit its protein expression. The overexpression of IASPP gene can inhibit the development of neurite and inhibit the effect of miR-124a on the growth of neurite. Conclusion miR-124a can promote the growth of neurite by using miR-124a. Inhibit the expression of iASPP gene to promote neurite growth.
