大鼠骨髓间充质干细胞向海绵体平滑肌细胞转化的实验研究

【摘要】: 目的: 观察在海绵体平滑肌细胞提取物诱导下,大鼠骨髓间充质干细胞(MSCs)向大鼠海绵体平滑肌细胞(CSMCs)转化的可能。 方法: 取4~5周左右SD大鼠,断颈处死后无菌条件下分离出股骨、胫骨。剪断干骺端后用5ml注射器吸取培养基冲洗骨髓腔,将液体直接冲入离心管中,用密度梯度离心法从冲洗出的细胞中分离出MSCs,进行原代培养。两代后,用不同数量CSMCs裂解获取的细胞提取物(cell extract, CE)进行诱导,24h后分别进行免疫组化分析并观察其形态学特性。 结果: 海绵体平滑肌细胞CE诱导的细胞均可表达α-actin,且高倍镜下观察可见部分海绵体平滑肌细胞特征,对照组中未见α-actin的表达,高倍镜下观察也无海绵体平滑肌细胞特征。CE组和对照组间有显著性差异,各CE组之间也有差异。即在海绵体平滑肌细胞CE诱导下,MSCs可以转化为CSMCs。 结论: MSCs在海绵体平滑肌细胞CE诱导下可以分化为CSMCs。
[Abstract]:Objective:. To observe the possible transformation of rat bone marrow mesenchymal stem cells (MSCs) into rat cavernous smooth muscle cells (CSMCs) induced by extractant from cavernous smooth muscle cells (SMC). Methods:. The femur and tibia were isolated from SD rats about 5 weeks after cervical dissection. After cutting off the metaphysis, the medullary cavity was washed with a 5 ml syringe and the liquid was washed directly into the centrifuge tube. MSCs were isolated from the washed cells by density gradient centrifugation and cultured in primary culture. After two generations, the cells were induced by different numbers of CSMCs cleavage cells (CES) for 24 hours and their morphological characteristics were analyzed by immunohistochemistry. Results:. Cavernous smooth muscle cells induced by CE could express 伪 -actinin. Some features of smooth muscle cells of cavernous body were observed under high power microscope, but no expression of 伪 -actin was found in control group. There was also no significant difference between CE group and control group under high power microscope, that is, MSCs could be transformed into CSMCsunder CE induced by cavernous smooth muscle cells. Conclusion:. MSCs can differentiate into CSMCs under CE induction of cavernous smooth muscle cells.
