霍乱弧菌肠毒素B亚单位基因在双歧杆菌中克隆及表达的初步研究

【摘要】:研究背景: 霍乱(Cholera)是一种由霍乱弧菌(Vibrio cholerae)引起的烈性传染病,一直以来都是各国传染病防治的重点,免疫接种是预防霍乱的最有效措施。以前的注射用疫苗不同程度存在着各种缺点,研究也显示针对霍乱这种肠道传染病的疫苗,通过口服途径给予的效果优于其他任何胃肠外给予途径,因为肠道粘膜免疫在霍乱的免疫预防中发挥着主要作用。因此,开发安全、方便、价廉的口服疫苗成为霍乱免疫预防研究的热点,而以霍乱弧菌肠毒素(Cholera toxin,CT)为基础的基因重组口服疫苗成为霍乱疫苗开发的主要方向。CT是霍乱弧菌主要的致病因子,它是分子量为84KD的肠毒素,由1个A亚单位(Cholera toxin Asubunit,CTA)和5个B亚单位(Cholera toxin B subunit,CTB)形成的五聚体共价连接而成,CTA为毒性亚单位,CTB为无毒的受体结合亚单位,后者是霍乱的主要保护性抗原。口服CTB可以刺激机体产生保护性粘膜抗体和血清抗体,发挥霍乱防治作用。此外,CTB还是一种很好的粘膜免疫佐剂,用于强化多糖、小肽等半抗原的免疫原性,与其他不相关抗原共同使用时,可以促进诱导其他抗原的粘膜免疫反应,,用于更广泛的人类疫苗的发展。所以,为充分发挥CTB的免疫特性作用,探索CTB在各种宿主表达的研究日益增多,如细菌、植物、
[Abstract]:Research background:
Cholera (Cholera) is a kind of Vibrio cholerae (Vibrio cholerae) caused by infectious diseases, has been the focus of the prevention and treatment of infectious diseases, vaccination is the most effective measure for the prevention of cholera. Previous vaccines had many defects in different degrees. This study also showed that for cholera intestinal infectious disease vaccine that effect was given via the oral route is better than any other way for parenteral administration, intestinal mucosal immunity plays a major role in the immune prevention of cholera. Therefore, the development of safe, convenient, inexpensive oral disease prevention research has become the focus of vaccine immunity to cholera, cholera toxin (Cholera toxin CT) gene recombinant oral vaccine based cholera vaccine development has become the main direction of.CT is the main pathogenic factor of Vibrio cholerae, which is a molecular weight of 84KD enterotoxin, composed of 1 A subunits (Cholera to Xin Asubunit, CTA) and 5 B (Cholera toxin B subunit subunit, CTB) five dimer formation of covalent connection, CTA is the toxic subunit, CTB is a non-toxic receptor binding subunit, which is the major protective antigen of cholera.
