【摘要】:背景: 长期摄入乙醇是引起男性不育发病率逐年增高的重要原因之一。而乙醇损害男性生殖能力的机制目前还未阐明,可能与睾酮的合成、分泌的减少有关。睾酮是机体始动和维持生精最重要内分泌调节激素,其合成和分泌主要受下丘脑-垂体-性腺轴调节。乙醇可以抑制下丘脑-垂体-性腺轴的活性,这是乙醇抑制睾酮合成和分泌的可能机制之一。近年来研究发现,乙醇还可直接作用于睾丸间质细胞,抑制睾酮的合成过程,并且作用环节位于睾酮合成的共同通路上,即线粒体合成过程。 PBR,外周型苯二氮卓类受体,在睾丸内有广泛表达,它主要分布在细胞内线粒体外膜上,被认为是胆固醇进入线粒体的控制阀。乙醇可影响多种组织PBR的表达:这与多种乙醇相关疾病的发生有关。乙醇是否会影响睾丸间质细胞PBR的表达,进而影响睾酮的线粒体合成过程目前还不清楚。 另外,PPARa(过氧化物酶体增生物活化受体a)是PBR表达的正调节因子,PPARa的激活剂可以显著上调大鼠肝脏PBR的表达。同时有研究发现乙醇可降调肝组织PPARa的表达,并且与酒精性脂肪肝的发生有关。上述研究进一步提示乙醇可通过影响PPARa,调节睾丸组织PBR的表达,进而影响睾酮合成的可能性。 ABP,即雄激素结合蛋白。是睾酮发挥生物学效应的重要的调节
[Abstract]:Background:. Long-term alcohol intake is one of the important reasons for the increasing incidence of male infertility. However, the mechanism of alcohol impairing male reproductive ability has not been elucidated, and may be associated with testosterone synthesis. Testosterone is the most important endocrine regulating hormone for initiation and maintenance of spermatogenesis. Its synthesis and secretion are mainly regulated by hypothalamus-pituitary-gonadal axis. Ethanol can inhibit the activity of hypothalamus-pituitary-gonadal axis. This is one of the possible mechanisms by which ethanol inhibits the synthesis and secretion of testosterone. In recent years, it has been found that ethanol also acts directly on the stromal cells of the testis and inhibits the synthesis of testosterone, which is located in the common pathway of testosterone synthesis. The process of mitochondrial synthesis. PBR, a peripheral benzodiazepine receptor, is widely expressed in the testis. Alcohol can affect the expression of PBR in many tissues: it is related to the development of many ethanol-related diseases. Whether ethanol affects the expression of PBR in the interstitial cells of testis. It is not clear that the mitochondrial synthesis of testosterone is affected. In addition, PPARAA (peroxisome proliferator-activated receptor a) is a positive regulator of PBR expression. The activator of PPARa can significantly up-regulate the expression of PBR in rat liver. At the same time, it has been found that ethanol can down-regulate the expression of PPARa in liver tissue. These results further suggest that ethanol can regulate the expression of PBR in testis and further influence the possibility of testosterone synthesis by affecting the expression of PBR in testis. ABP, or androgen-binding protein, is an important regulation of testosterone's biological effects.
