斯氏狸殖吸虫对大鼠肝脏损害的实验研究

【摘要】:【目的】研究斯氏狸殖吸虫对大鼠肝脏结构和功能的损害情况;探讨斯氏狸殖吸虫引起大鼠肝脏损害的机制;评价肝功能检查在斯氏狸殖吸虫病肝脏损害的早期诊断中的价值。 【方法】 35只SD大鼠随机分成3组:对照组、实验A组、实验B组,将斯氏狸殖吸虫囊蚴经腹腔注射感染实验组大鼠以建立动物模型(每鼠注射15个囊蚴),用全自动生化分析仪于感染后不同时间分别检测对照组和实验A组大鼠血清总蛋白、白蛋白、球蛋白、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、血清总胆红素、血清结合胆红素与非结合胆红素等肝功能指标,并进行统计分析;于感染后不同时间分别剖杀实验B组的部分大鼠,用光学显微镜、透射电镜观察肝脏组织病理学变化。 【结果】斯氏狸殖吸虫对大鼠肝脏造成明显损害,肝脏功能损害主要表现为:感染后4、6、8周实验A组大鼠血清白蛋白含量明显下降,感染后4、6、8、52周血清球蛋白含量明显升高,而总蛋白含量在感染后变化不明显;感染后6、8周,ALT与AST活性均升高;血清总胆红素、血清结合胆红素与非结合胆红素在感染后改变不明显;肝功能各指标在感染后的前2周变化不明显。肝脏结构损害主要表现为:感染后1周,肝细胞水肿,少数肝细胞脂肪变性;感染后2周,肝脏突出的病变为急性化脓性炎症,在肝组织内形成小脓肿;感染后8周,肝组织内出现片状纤维组织增生,部分肝小叶结构破坏、重建:感染后52周,肝脏纤维化改变明显,有大量假小叶形成。透射电镜观察到:从感染后2周开始,实验B组大鼠肝脏均呈现出不同程度的纤维化,8周以后纤维化明显
[Abstract]:[objective] to study the damage of paragonimus skrjabini on the structure and function of rat liver, to explore the mechanism of hepatic damage induced by P. skrjabini, and to evaluate the value of liver function examination in the early diagnosis of hepatic damage caused by paragonimiasis skrjabini. [methods] Thirty-five Sprague-Dawley rats were randomly divided into three groups: control group, experimental group A, experimental group B. In order to establish the animal model, the rats of experimental group were infected with Cysticercus skrjabini by intraperitoneal injection. The serum total protein of control group and experimental group A were detected by automatic biochemical analyzer at different time after infection. Albumin, globulin, alanine aminotransferase (alt), aspartate aminotransferase (AST), serum total bilirubin, serum conjugated bilirubin and unconjugated bilirubin were analyzed statistically. Some rats in group B were killed at different time after infection. The histopathological changes of liver were observed by optical microscope and transmission electron microscope. [results] the liver of rats was obviously damaged by paragonimus skrjabini. The main damage of liver function was as follows: the serum albumin content of experimental group A decreased significantly, and the content of serum globulin increased significantly at 852 weeks after infection in group A. The serum total bilirubin, serum conjugated bilirubin and unconjugated bilirubin did not change significantly after infection. The changes of liver function in the first two weeks after infection were not obvious. The main structural damage of liver was: 1 week after infection, edema of liver cells, fatty degeneration of a few liver cells, and acute suppurative inflammation 2 weeks after infection. Small abscess was formed in the liver tissue, and at 8 weeks after infection, the flake fibrous tissue proliferated and some hepatic lobules were destroyed, and the liver fibrosis changed obviously at 52 weeks after infection. A large number of pseudolobules were formed. It was observed by transmission electron microscope that from 2 weeks after infection, the liver of experimental group B showed various degrees of fibrosis.
