一次免疫神经氨酸酶DNA疫苗抗流感病毒研究

【摘要】:流感是一种烈性传染性疾病,每年造成很多人的死亡。DNA疫苗作为一种前景广阔的疫苗已广泛应用于流感病毒预防研究。本论文分三个部分介绍我们在DNA疫苗研究方面的工作: Ⅰ.我们以前的研究表明NA、HA DNA疫苗在小鼠模型上诱导的免疫反应可以提供抗流感病毒保护,但是一般需要给小鼠接种两次NA或HA DNA疫苗才能够完全保护。为了简化免疫程序,本研究采用了一次免疫策略来检测NA、HA DNA疫苗的免疫效果。我们用电穿孔的方法给小鼠接种一次NA DNA或HA DNA,检测其保护小鼠的能力。BALB/c小鼠接种一次30μg NA、HA,在小鼠免疫后的第4周、第28周分别用致死量(40×LD_(50))流感病毒攻击。实验表明接种一次30μg NA不但能在免疫后4周保护小鼠,而且在免疫后28周仍然能够保护。接着,我们又对接种一次NA DNA进行了剂量研究。实验表明,小鼠接种5μg NA就能在致死量流感病毒攻击下存活,接种了低于这个剂量的小鼠在攻击后1周全部死亡。最后,我们检测了NA DNA在新生小鼠体内的保护作用,结果发现,一日龄和一周龄小鼠免疫了30μg NA后在致死量病毒攻击下存活数量具有显著的统计学意义。在以上实验中接种一次30μg HA的小鼠在攻击后的存活数都没有统计学意义。综合以上实验结果,NA作为一种简单有效的DNA疫苗,对预防流感有积极意义。 Ⅱ.本部分旨在以小鼠为动物模型,研究母体免疫流感病毒血凝素(hemagglutinin,HA)DNA疫苗对子代小鼠的保护作用。在本实验中,雌
[Abstract]:Influenza is a severe infectious disease. As a promising vaccine, DNA vaccine has been widely used in influenza virus prevention research. This paper introduces our work on DNA vaccine research in three parts:. I. Our previous studies have shown that the immune response induced by NAHA-HA DNA vaccine in mice can provide anti-influenza virus protection, but it usually takes twice to vaccinate mice with na or HA DNA to be fully protected. In this study, we used one time immunization strategy to detect the immune effect of NAA HA DNA vaccine. We used electroporation method to inoculate mice with na DNA or HA DNA once. The ability of protecting mice. BALB- c mice were inoculated with 30 渭 g NAA at 4 weeks after immunization. The experiment showed that inoculation with 30 渭 g na could protect mice not only 4 weeks after immunization, but also 28 weeks after immunization. We also carried out a dose study of na DNA inoculation. The experiment showed that mice inoculated with 5 渭 g na could survive the lethal influenza virus attack, and all the mice inoculated with less than this dose died one week after the attack. We examined the protective effect of na DNA in newborn mice and found that, The number of mice immunized with 30 渭 g na for one day and one week after inoculation with lethal viral attack was statistically significant.
