皮质颗粒在评估体外受精中卵母细胞胞质成熟度作用的研究

【摘要】:近年来一些研究认为卵母细胞胞核和胞质成熟程度,尤其是胞质成熟度是影响体外受精—胚胎移植(in vitro fertilization-embryo transfer,IVF-ET)成功率的一个重要原因。卵母细胞成熟度包括胞核成熟和胞质成熟,第一极体(first body,PB1)是明显的可以直接观察到的细胞核成熟的特征,而如何观察和评价卵母细胞胞质成熟程度,进而分析受精失败的原因,一直是我们面临的难题;目前大多数学者把皮质颗粒(cortical granule,CG)的分布和迁移视为评价胞质成熟度的一项重要指标。CG的皮质反应和透明带反应是保证正常受精和阻止多精受精的两个重要生理功能;CG的这些特性,既是细胞质成熟的一个标志,也是细胞质成熟的重要部分;所以本研究将CG在卵母细胞质膜下的线性排列作为细胞质成熟的指标。关于人卵母细胞CG的分布情况,国内尚未见到报道。本研究以CG作为评价胞质成熟的指标,一方面以昆明小白鼠为模型,研究了卵母细胞成熟过程中CG的迁移和排放情况,证实了CG的迁移及分布情况与卵母细胞的成熟度有着直接的关系,在保证正常受精及阻止多精受精方面发挥了重要作用;另一方面通过荧光显微镜观察了人IVF周期中未成熟、未受精、多精受精卵及发育阻滞的合子中CG的分布和迁移情况,对卵母细胞进行了胞质成熟度的评估。本研究旨在快速判断卵母细胞的成熟度,找出常规IVF第一周期受精率低或受精失败的原因,为患者以后的治疗提供基础与临床上的理论依据,从而协助患者选择下一周期的最佳治疗方法、受精方式和体外培养体系。
[Abstract]:In recent years, some studies have suggested that oocyte nuclear and cytoplasmic maturation, especially cytoplasmic maturity, is an important factor affecting the success rate of in vitro fertilization and embryo transfer in vitro fertilization-embryo transfer IVF-ET.The maturation of oocytes includes nuclear maturation and cytoplasmic maturation. The first polar body first body (PB1) is an obvious characteristic of nuclear maturation which can be observed directly. However, how to observe and evaluate the cytoplasmic maturation of oocytes and then analyze the causes of fertilization failure has been a difficult problem. At present, most scholars regard the distribution and migration of cortical granuleus (CGG) as an important index for evaluating cytoplasmic maturity. CG's cortical reaction and pellucida response are two important physiological tasks to ensure normal fertilization and prevent multiple fertilization. These are the properties of CG, It is not only a marker of cytoplasmic maturation, but also an important part of cytoplasmic maturation. In this study, the linear arrangement of CG under oocyte plasma membrane was taken as the index of cytoplasmic maturation. In this study, CG was used as an index to evaluate cytoplasmic maturation. On the one hand, the migration and emission of CG during oocyte maturation were studied using Kunming mice as a model. It was proved that the migration and distribution of CG was directly related to the maturation of oocytes, which played an important role in ensuring normal fertilization and preventing poly-spermatid fertilization, on the other hand, the immature of human IVF cycle was observed by fluorescence microscope. The distribution and migration of CG in unfertilized, polysperm fertilized eggs and developmental retarded zygotes were evaluated to evaluate the cytoplasmic maturity of oocytes. To find out the causes of low fertilization rate or failure of fertilization in the first cycle of conventional IVF, to provide basic and clinical theoretical basis for the subsequent treatment of patients, so as to assist patients to select the best treatment method, fertilization method and in vitro culture system for the next cycle.
