tat蛋白提高HIV-1疫苗免疫水平的研究

【摘要】:DNA 疫苗研究的一个重要任务是如何能更好地提高DNA 疫苗的体液免疫和细胞免疫。近年来的研究显示,HIV 的转录活化因子—Tat 蛋白具有自由通过细胞质膜的特性。现已有不少借助Tat 蛋白的穿膜活性将其它蛋白或DNA 成功导入活细胞内的报道。根据大量文献提供的理论基础设计了实验,将tat 与gag/pol 融合表达,利用Tat 蛋白的穿膜活性将gag/pol 蛋白导入其它活细胞内,从而提高DNA 疫苗免疫水平。还有研究表明Tat49-57 短肽T9(RKKRRQRRR)有较强的穿膜功能,因此我们将这T9 与gag/pol融合表达,研究T9 的插入对DNA 疫苗的免疫水平的影响。 本论文的主要工作是构建DNA 疫苗,将tat、T9 与HIV-1gag/pol 融合表达,检测DNA 疫苗的抗原性和免疫原性,研究Tat、T9 蛋白对提高HIV-1 DNA 疫苗免疫水平的作用。实验结果证明tat和gag/pol 融合表达能提高DNA 疫苗的细胞免疫水平。Tat 基因的插入提高HIV-1 DNA 疫苗的有效性,也为tat 应用于其他疫苗中提供了广阔实际应用前景。 实验中我们发现T9 与gag/pol 的融合表达并不是象我们预期设想那样能提高HIV-1 DNA 疫苗的免疫水平,反而有所降低,我们将在以后的实验中进一步分析研究其原因。
[Abstract]:One of the important tasks of DNA vaccine research is how to improve the humoral and cellular immunity of DNA vaccine. Recent studies have shown that the transcriptional activator Tat protein of DNA has the property of free passing through the cytoplasmic membrane. There are many reports that other proteins or DNA have been successfully introduced into living cells by the transmembrane activity of Tat protein. Experiments have been designed based on the theoretical basis provided by a large number of literatures. The expression of tat and gag/pol was fused, and the gag/pol protein was introduced into other living cells by the transmembrane activity of Tat protein, so as to improve the immune level of DNA vaccine. Studies also showed that Tat49-57 short peptide T9 RKKRRQRRRRR) had strong transmembrane function, so we fused the expression of T9 with gag/pol. To study the effect of T 9 insertion on the immune level of DNA vaccine. The main work of this thesis is to construct DNA vaccine, to express T9 with HIV-1gag/pol, and to detect the antigenicity and immunogenicity of DNA vaccine. To study the effect of Tatt T9 protein on improving the immune level of HIV-1 DNA vaccine. The results show that the fusion expression of tat and gag/pol can improve the cellular immunity level of DNA vaccine. Tat gene insertion can improve the effectiveness of HIV-1 DNA vaccine. It also provides a broad prospect for the application of tat in other vaccines. In our experiment, we found that the fusion expression of T9 and gag/pol did not increase the immune level of HIV-1 DNA vaccine as we expected, but decreased, and we will further analyze and study its cause in future experiments.
