人早期心脏发育过程中VEGF及其受体的表达研究

【摘要】:目的:血管内皮生长因子(vascular endothelial growth factor, VEGF)家族是由细胞分泌的多肽及蛋白质类化学信息分子,也是血管生成刺激因子中最重要的一类。许多研究证实新血管的生成和发育与VEGF家族有密切联系。本实验第一次采用人胚做材料,研究人胚心发生、发育的过程以及VEGF-A、VEGF-C、VEGFR-1、VEGFR-2、VEGFR-3在第3~8周人胚心内皮细胞、成心肌细胞中的表达情况,以进一步探讨VEGF-A和VEGF-C在人早期心发生、发育过程中所起的作用。 材料及方法:收集意外流产、受精龄为3个月内的新鲜人胚。固定、包埋、切片、观察,,根据胚层形成情况、体节的数目及器官发育状况挑选胚龄为3~8w的标本切片。采用HE染色观察早期心的外形及心内皮细胞、成心肌细胞的形态学变化;免疫组化对VEGF-A和VEGF-C及受体VEGFR-1、VEGFR-2、VEGFR-3在心内皮细胞及成心肌细胞的表达情况进行研究。 结果:HE染色结果显示人胚第18、19天,脏壁中胚层内的成血管细胞局部增生形成生心区(cardiogenic area)。第3周末,形成纵行排列的内
[Abstract]:Objective: vascular endothelial growth factor (VEGF) family is a family of polypeptide and protein molecules secreted by cells. It is also one of the most important angiogenic stimulators. Many studies have shown that the formation and development of new blood vessels are closely related to the VEGF family. The process of development and the expression of VEGFR-1 and VEGFR-2VEGFR-3 in human embryonic heart endothelial cells and cardiomyocytes at the 3rd week were discussed in order to further explore the role of VEGF-A and VEGF-C in the early development of human heart. Materials and methods: fresh human embryos with accidental abortion and fertilization age within 3 months were collected, fixed, embedded, sectioned, observed, and based on the formation of the embryo. The number of somatic ganglia and organ development were selected for 8 weeks of embryo. The morphological changes of cardiac endothelial cells and cardiomyocytes were observed by HE staining. The expression of VEGF-A and VEGF-C and VEGFR-2 and VEGFR-3 in cardiac endothelial cells and cardiomyoblasts were studied by immunohistochemistry. Results on the 1819 th day of human embryo, the vascular cells in the mesoderm of the visceral wall were locally proliferated to form cardiogenic area cardiogenic area. At the end of the third week, they formed a longitudinal arrangement.
