【摘要】:目的:鉴定酸性肽(acidic peptide,AP)在动物脑中是否普遍存在,测定大鼠脑内酸性肽和谷氨酸(glutamic acid,Glu)的含量,并探讨外源性酸性肽对大鼠内源性酸性肽及谷氨酸含量的影响。 方法:用纸电泳法(300V,1h)分离定位marker,此marker包括谷氨酸、酸性肽和谷胱甘肽三种标准小肽或氨基酸,然后用此marker鉴别牛、羊、鸡、鸭、兔、大鼠等多种动物脑中AP的存在。用标准谷氨酸、酸性肽和谷胱甘肽配成浓度分别为0.5mg/ml、1.0mg/ml、2.0mg/ml、3.0mg/ml、4.0mg/ml的混合液,每一浓度各取4μl点样,然后进行电泳分离,在烘箱中烘10min,剪下斑点,洗脱液洗脱,分光光度法测定不同浓度的AP和Glu的吸光度值(OD)。重复10次实验。以平均OD值为纵坐标,浓度或含量为横坐标,绘制AP和Glu的标准曲线。将大鼠随机分为6组(每组10只),其中1组为对照组,其余5组按100mg/kg酸性肽灌胃,分别在灌胃后1h、
[Abstract]:Aim: to determine the prevalence of acidic peptide (APD) in animal brain, to determine the contents of acidic peptide and glutamic acid acid acid (glutamic acid acid) in rat brain, and to investigate the effect of exogenous acidic peptide on the content of endogenous acid peptide and glutamate in rats. Methods: Marker was isolated by paper electrophoresis for 1 h. The marker consisted of glutamic acid, acid peptide and glutathione. The marker was used to identify cattle, sheep, chicken, duck and rabbit. The presence of AP in the brains of rats and other animals.
