脏器间质树突状细胞在多器官功能障碍综合征中的变化与作用研究

【摘要】:目的:观察小鼠肺脏、肝脏与肾脏间质树突状细胞的形态、数量和分布,了解MODS小鼠肺脏、肝脏与肾脏间质树突状细胞病变以及DC功能相关的细胞因子、趋化因子及炎症因子在MODS病程中的变化规律与意义,探讨间质DC在免疫紊乱、过度炎症及脏器损伤机制中的影响与作用,为MODS免疫学机制与防治研究提供新的依据。 方法:腹腔注射酵母多糖复制C57BL/6小鼠MODS模型,分为正常对照、3-6小时、12-48小时、5-7日和10-12日组。光镜与电镜观察MODS各期小鼠肺脏、肝脏和肾脏组织病理学改变,脏器间质树突状细胞的形态、数量和分布特点及变化规律。运用免疫学方法检测间质树突状细胞表面标记物CD11c和CD205、共刺激分子CD80、CD86和MHC-Ⅱ类分子的表达水平及肺脏中IL-1β、IL-10及CGRP的表达情况。运用RT-PCR法检测趋化因子SLC及其受体CCR7在肺脏中的表达变化情况,判定MODS各期肺间质树突状细胞的迁移活性。流式细胞术检测MODS各期外周血CD4~+T细胞与CD8~+T细胞的数量与比值,外周血单个核细胞对MHC-Ⅱ分子表达的变化。 结果:间质树突状细胞广泛分布于正常小鼠肺脏、肝脏与肾脏中,数量极少,约占各脏器总细胞数的5-9‰,大部分处于非成熟状态,低表达CD80、CD86与MHC-Ⅱ分子。致伤早期(3-6小时),各脏器功能与病理改变轻微。间质DC显著增生,对CD80、CD86和MHC-Ⅱ分子的表达轻度增加。外周血中MHC-Ⅱ阳性的单个核细胞与CD4~+T细胞数均减少,CD4/8比值下降。趋化因子SLC及其受体CCR7在肺组织中表达水平均较正常组轻度上升。炎症因子IL-1β、IL-10和CGRP在肺组织中阳性细胞数较正常组显著升高;急性损伤期(12-48小时),各脏器出现功能改变,脏器实质细胞损伤较重。间质DC继续保持增生状态,数量略低于致伤早期,但CD80、CD86和MHC-Ⅱ
[Abstract]:Objective: to observe the morphology, quantity and distribution of interstitial dendritic cells in lung, liver and kidney of mice, and to understand the cytokines related to the function of DC in lung, liver and kidney of MODS mice. The changes of chemokines and inflammatory factors in the course of MODS were studied. The effects of interstitial DC on immune disorder, excessive inflammation and organ injury were discussed, which provided a new basis for the study of immunological mechanism and prevention and treatment of MODS. Methods: the MODS model of C57BL/6 mice was induced by intraperitoneal injection of yeast polysaccharide. The mice were divided into normal control group (3-6 hours, 12-48 hours, 5-7 days and 10-12 days). The histopathological changes of lung, liver and kidney of MODS mice were observed by light and electron microscopy.
