一氧化氮在酒精抑制大鼠离体结肠运动中的介导作用及其机制研究

【摘要】:饮酒在东、西方国家都很常见,酒精中毒的副效应也被人们广泛关注。急性饮酒对消化道的主要副作用包括:恶心、呕吐和腹泻。已报道酒精能够抑制胃肠道运动,但其抑制效应的机制还不清楚,酒精对结肠运动有何影响,也未见文献报道。 一氧化氮(Nitric Oxide,NO)作为胃肠道内非肾上腺非胆碱能神经(nonadrenergic noncholinergic,NANC)的主要抑制性神经递质,,能够诱导食管、小肠、大肠等部位的平滑肌舒张。NO也能调节乙酰胆碱、血管活性肠肽、P物质等配体对血管平滑肌的舒张作用;NO可能也介导了酒精对食管下端括约肌运动的抑制。NO是否介导酒精对消化道其他部位运动的抑制目前未见报道。 女性和雌性动物对酒精的敏感性与男性和雄性动物相比有明显的不同,我们前期研究也发现腹腔注射酒精会引起胃排空和小肠推送活动减弱,且雌性动物的反应明显比雄性强。目前尚不清楚酒精对结肠的作用是否也存在性别差异。 实验选用健康成年Wistar大鼠,实验前禁食12小时。制备大鼠离体结肠近端纵行肌(LP)和结肠近端环行肌(CP)肌条,将肌条置入盛有5ml Krebs液(37℃)并充有95%O_2和5%CO_2混合气体的灌流肌槽中孵育,记录其自发收缩活动。以肌条收缩曲线下面积(即张力积分)为指标,观察酒精(2.0mL/L、3.3mL/L、5.0mL/L、
[Abstract]:Alcohol consumption is common in eastern and western countries, and the side effects of alcoholism are widely concerned. The main side effects of acute drinking on the digestive tract include nausea, vomiting and diarrhoea. It has been reported that alcohol inhibits gastrointestinal motility. However, the mechanism of its inhibitory effect is unclear, and the effect of alcohol on colon motility has not been reported. Nitric oxide nitric oxide (no) is the main inhibitory neurotransmitter of nonadrenergic noncholinergic noncholinergic NANCin the gastrointestinal tract, which can induce relaxation of smooth muscle in esophagus, small intestine, large intestine and so on. No can also regulate acetylcholine. Relaxation of vascular smooth muscle by ligand such as vasoactive intestinal peptide substance P no may also mediate the inhibition of lower esophageal sphincter movement by alcohol. Whether no mediates the inhibition of alcohol on other parts of digestive tract has not been reported. The sensitivity of women and females to alcohol is significantly different from that of males and males. Previous studies have also found that intraperitoneal injection of alcohol leads to reduced gastric emptying and small bowel push. The response of female animals is significantly stronger than that of males. It is not clear whether there are gender differences in the effects of alcohol on the colon. The healthy adult Wistar rats were fasted for 12 hours before the experiment. The muscle strips of isolated proximal longitudinal muscle of colon and proximal circular muscle of colon were prepared in this experiment.
