ConA的胰蛋白酶消化产物对培养家兔静息软骨细胞的增殖分化影响

【摘要】:变形性关节炎等退形性关节疾病和关节机械性损伤的发病率在逐年提高。由于软骨组织结构的特殊性,缺乏血管,淋巴,神经系统的营养供给,给得软骨疾病的治疗恢复带来了很大的困难。应用组织工程学技术体外构建人工软骨,尝试关节软骨损伤的修复是国际上该领域的研究热点,软骨细胞也是最早应用于组织工程的种子细胞之一,但由于软骨细胞作为种子细胞,其来源困难,增殖能力和数量有限,体外长期培养的软骨细胞易发生反分化现象,失去软骨细胞的生物活性,限制了其应用。因此对软骨的增殖分化机制的研究,寻找强效的刺激因子必然对软骨组织工程的发展,软骨疾病的防治带来很大的帮助。 刀豆球蛋白(Concanavalin A,简称ConA)是Sumner 1919年首次从大刀豆中分离纯化出来的一种植物凝集素,后来Sumner和Howell[19]发现纯化的ConA不仅能凝集红细胞,而且还能沉淀溶液中的糖元和淀粉。随着ConA的研究的深入,ConA的生物活性也逐渐得到认识,ConA对淋巴细胞具有强烈的促有丝分裂作用,刺激
[Abstract]:The incidence of degenerative joint diseases and mechanical joint injuries such as deformable arthritis is increasing year by year. It is very difficult to treat and recover the cartilage diseases. It is a hot spot in the world to construct artificial cartilage in vitro and try to repair the articular cartilage injury by using tissue engineering technology. Chondrocytes are also one of the first seed cells used in tissue engineering. However, because of the difficulty of chondrocytes as seed cells, their proliferation ability and quantity are limited, the long-term cultured chondrocytes are prone to dedifferentiation. The loss of the biological activity of chondrocytes limits its application. Therefore, the study on the mechanism of cartilage proliferation and differentiation and the search for strong stimulating factors are bound to be of great help to the development of cartilage tissue engineering and the prevention and treatment of cartilage diseases. Concanavalin A (Cona) is the first plant agglutinin isolated from concanavalin by Sumner in 1919. Later, Sumner and Howell [19] found that the purified ConA not only agglutinated red blood cells, Along with the further study of ConA, the biological activity of Cona has been gradually recognized that Cona has a strong mitogenic effect on lymphocytes and stimulates it.
