球形幽门螺杆菌对胃上皮细胞增殖与凋亡及细胞内转录因子Egr-1表达的影响

【摘要】: 幽门螺杆菌(Helicobacter pylori, Hp)在营养不足,抗生素处理等胁迫因素作用下,其形态会发生适应性转变,由螺旋形转变为球形。球形幽门螺杆菌具有免疫原性低,不易清除等特点。研究球形幽门螺杆菌对宿主细胞增殖与凋亡的影响,以及对宿主细胞内转录因子Egr-1表达的影响,将有助于了解球形Hp感染与各种胃病特别是胃癌发生的关系,从细胞微生物学的角度深入探讨幽门螺杆菌的致病机理。 一、球形Hp对胃上皮细胞增殖与凋亡的影响 目的:了解球形幽门螺杆菌对人胃上皮细胞增殖与凋亡的影响。方法:选择正常胃上皮细胞株GES-1和胃癌细胞株AGS为研究对象,将球形和螺旋形Hp分别与它们共培养。利用CCK-8法检测活细胞的细胞增殖情况,用TUNEL法检测细胞凋亡情况。并比较在相同感染密度下,两种形态的Hp对胃上皮细胞的增殖与凋亡的作用强度是否存在差异。结果:1.在低感染密度状态下,球形Hp的促细胞增殖作用高于对照组(PI=2.6%~5.0%),P0.05。当MOI=32:1时,球形Hp处理AGS细胞OD值为2.902±0.072,细胞增殖率(PI)62.58%。明显高于螺旋形Hp处理组(PI=18.04%),P0.05。当MOI=16:1时,球形Hp处理GES-1细胞OD值为1.808±0.156,细胞增殖率32.07%,明显高于螺旋形Hp处理组(PI=21.91%),P0.05。2.在低感染密度状态下,球形Hp的诱导凋亡作用较弱,与对照组(AI=3.24%~5.42%)相比,无明显差异,P0.05。当MOI=32:1时,球形Hp处理的AGS细胞,凋亡率(AI)为5.94%,明显低于螺旋形Hp处理组(AI=14.52%),P0.05。当MOI=16:1时,球形Hp处理GES-1细胞,凋亡率为6.81%,明显低于螺旋形Hp处理组(AI=16.25%),P0.05。结论:1.在Hp的低感染密度状态下,与螺旋形Hp相比,球形Hp对胃上皮细胞表现出较强的促增殖作用和相对较弱的诱导凋亡的作用。故球形Hp对体外胃上皮细胞具有较强的促增殖效应。2.球形Hp对正常胃上皮细胞和胃癌细胞增殖与凋亡的影响基本一致。 二、球形Hp刺激AGS细胞增殖过程中转录因子Egr-1的作用 目的:活化的Egr-1在胃上皮细胞的恶性转化中具有重要作用。检测球形Hp对胃
[Abstract]:The morphology of Helicobacter pylori (HP) changed from spiral to spherical under stress factors such as inadequate nutrition and antibiotic treatment.Helicobacter pylori globular has low immunogenicity and is difficult to be cleared.To study the effects of Helicobacter pylori on the proliferation and apoptosis of host cells and on the expression of transcription factor Egr-1 in host cells will contribute to the understanding of the relationship between spherical HP infection and the occurrence of various gastric diseases, especially gastric cancer.To explore the pathogenic mechanism of Helicobacter pylori from the point of view of cell microbiology.1. Effects of spherical HP on proliferation and apoptosis of gastric epithelial cellsObjective: to investigate the effect of Helicobacter pylori on proliferation and apoptosis of human gastric epithelial cells.Methods: normal gastric epithelial cell line (GES-1) and gastric cancer cell line (AGS) were selected and cocultured with spherical and spiral HP respectively.CCK-8 assay was used to detect cell proliferation and TUNEL assay was used to detect cell apoptosis.At the same infection density, the effects of HP on gastric epithelial cell proliferation and apoptosis were compared.The result is 1: 1.Under the condition of low infection density, the proliferative effect of globular HP was higher than that of the control group (PI2.6and 5.0P0.05).When treated with MOI=32:1, the OD value of AGS cells treated with spherical HP was 2.902 卤0.072, and the cell proliferation rate was 62.58.It was significantly higher than that of helical HP treatment group (P 0.05).When treated with MOI=16:1, the OD value of GES-1 cells treated with spherical HP was 1.808 卤0.156, and the cell proliferation rate was 32.07, which was significantly higher than that of helical HP treatment group (P _ I _ (21.91)) and P _ (0.05.2).Under the condition of low infection density, the apoptosis-inducing effect of globular HP was weaker than that of the control group (AI3.24 and 5.42), there was no significant difference between them (P0.05).When treated with MOI=32:1, the apoptotic rate of AGS cells treated with spherical HP was 5.94%, which was significantly lower than that of spiral HP treatment group (14.52% P0.05).When treated with MOI=16:1, the apoptotic rate of GES-1 cells treated with spherical HP was 6.81, which was significantly lower than that of helical HP treatment group (P 0.05).Conclusion 1.Under the condition of low infection density of HP, compared with helical HP, spherical HP could promote the proliferation and induce apoptosis of gastric epithelial cells.Therefore, spherical HP has a strong proliferative effect on gastric epithelial cells in vitro.The effects of spherical HP on the proliferation and apoptosis of normal gastric epithelial cells and gastric cancer cells were consistent.Second, the role of transcription factor Egr-1 in the proliferation of AGS cells stimulated by spherical HPObjective: activated Egr-1 plays an important role in malignant transformation of gastric epithelial cells.Detection of spherical HP on stomach
