精液对子宫内膜蜕膜化及胚胎发育的影响

【摘要】:目的:探讨精液成分对子宫内膜蜕膜化及胚胎发育的影响。 方法:1.分离育龄期妇女增殖晚期及分泌期子宫内膜细胞,加入雌二醇(E2)、孕酮(P)使其蜕膜化,分别加入人精子及精浆共培养,观察蜕膜化的间质细胞生长情况并测定培养液中泌乳素PRL 含量。2.内膜培养8 天后,加入小鼠2-细胞期胚胎继续培养,观察胚胎的发育情况及内膜细胞生长情况。3.建立假孕鼠模型,观察精液在胚胎移植中的作用。 结果:1.加入精浆的子宫内膜间质细胞蜕膜化程度明显优于精子组和对照组。精浆组分泌PRL 的量与精子组和对照组相比有显著性差异(p0.05),而精子组和对照组无明显差异。2.精浆能促进八细胞以后的胚胎在子宫内膜细胞上的卵裂、囊胚形成、孵化、黏附和扩展性生长(p0.05)。3. 胚胎移植过程中精子对植入率没有影响。 结论:1 精浆对子宫内膜间质细胞蜕膜化有明显促进作用,能够促进PRL 的分泌。2.精浆的存在可以促进胚胎在子宫内膜细胞共培养体系中的发育。3.以输卵管结扎和假孕鼠为受体的胚胎移植着床率无明显差异提示了精浆与精液对囊胚着床作用可能相同。
[Abstract]:Objective: to investigate the effect of semen composition on decidualization and embryo development of endometrium.Method 1: 1.Endometrial cells of late proliferative and secretory phase were isolated from women of childbearing age. The decidua cells were treated with estradiol E2P and progesterone. The cells were co-cultured with human spermatozoa and seminal plasma, respectively.The growth of decidualized mesenchymal cells was observed and the content of prolactin PRL in culture medium was determined.After 8 days of endometrial culture, mouse 2-cell embryos were added to continue to culture, and the development of embryo and the growth of endometrial cells were observed.The role of semen in embryo transfer was observed by establishing pseudopregnant mouse model.The result is 1: 1.The degree of decidualization of endometrial stromal cells added seminal plasma was better than that of spermatozoa group and control group.The amount of PRL secreted in seminal plasma group was significantly different from that in spermatozoa group and control group (p0.05), but there was no significant difference between spermatozoa group and control group.Seminal plasma can promote the cleavage, blastocyst formation, hatching, adhesion and extensibility of the embryo in endometrial cells.Spermatozoa have no effect on implantation rate during embryo transfer.Conclusion the seminal plasma of 1: 1 can promote decidualization of endometrial stromal cells and promote the secretion of PRL.The existence of seminal plasma can promote the development of embryo in the co-culture system of endometrial cells.There was no significant difference in implantation rate between oviduct ligation and pseudopregnant mice, suggesting that seminal plasma and semen might have the same effect on blastocyst implantation.
