禽流感核酸疫苗免疫保护性研究

【摘要】:本论文由二部分组成(一)DNA疫苗与灭活疫苗混合免疫和单独免疫不同的动物能提供抗H5N1亚型禽流感病毒的保护性研究(二)血凝素(hemagglutinin,HA)和神经氨酸酶(neuraminidase,NA)DNA免疫BALB/c小鼠抗H9N2亚型禽流感病毒的保护性研究。 (一) 高致病性H5N1型禽流感病毒在1997年第一次突破种属障碍传给人,造成至少18例感染,6人死亡。禽流感病毒还仍然活跃在各国家禽市场,并影响世界的经济秩序和威胁到人类的生命安全。因此,必须预先接种疫苗诱导保护性免疫。本实验分别以6—8周龄BALB/C小鼠和三黄鸡为实验动物,分四组免疫:第一组,全病毒灭活疫苗初免,HA-DNA加强:第二组HA-DNA初免,灭活疫苗加强;第三组灭活疫苗免疫二次,第四组HA-DNA免疫二次,间隔三周,加强免疫后三周,用致死量(20×LD_(50))的H5N1亚型禽流感病毒(A/Chichen/JiangSu/01/2002)攻击小鸡和小鼠,测定动物血清抗体变化、动物的存活率及攻毒后第三天小鸡泄殖腔的排毒量和小鼠肺部的病毒残存量。结果表明四种免疫方式能为不同的动物提供抗H5N1型禽流感病毒保护和交叉保护。(二) H9N2型禽流感病毒广泛分布于禽类中,并且在香港和广东地区的人类感染者体内能够分离得到。我们应当寻找有效的途径控制H9N2型禽流感给人类带来的危害。最近分离的A/Chichen/JiangSu/07/2002(H9N2)病毒经肺对肺传代能使BALB/C小鼠致死,表明H9N2型禽流感病毒能在小鼠肺部大量扩增。我们以BALB/C小鼠为动物模型,分别用3μg、10μg、30μgHA-DNA、NA-DNA电击免疫6-8周龄的BALB/C小鼠一次或二次,一次免疫组于初免后四周用致死量(40×LD_(50))的H9N2型病毒攻击。二次免疫组间隔三周后加强免疫,加强免疫后一周用致死量(40×LD_(50))
[Abstract]:This paper consists of two parts (one is mixed immunization with inactivated vaccine and the other is mixed immunization with inactivated vaccine) and the other is to immunize BALB/c mice with H5N1 subtype Avian Influenza virus.
