人眼直肌Pulley系统组织形态学的实验研究

【摘要】:目的 采用组织化学及免疫组织化学技术、全眼眶连续冠状切片技术、计算机辅助三维重建技术研究人眼直肌Pulley的组织构成及其在眼眶内的立体空间分布情况,从多方面、多角度展示Pulley的组织形态结构,为探讨Pulley功能提供解剖学和组织学依据。 方法 (1) 选取5例无眼部疾患的新鲜尸体的眼眶,整体固定包埋,进行全眼眶连续冠状切片。相邻层面经Masson和Weigert两种特殊染色及免疫组织化学染色后,从大体角度和在光镜下观察每个层面的组织结构。 (2) 选取Pulley及Pulley连接带典型的组织切片层面经苦味酸一天狼星红染色后,在偏振光显微镜下观察直肌Pulley及Pulley连接带的胶原组成成分及分布特点。并用Mias-2000图像分析系统分析四条直肌Pulley及Pulley连接带的胶原组成。 (3) 经Masson染色的全眼眶冠状位连续组织切片图像用数码相机摄像后逐一输入计算机内,经photoshop 7.0软件除噪处理,应用3D-Doctor软件通过对图像的定位、分割进行Pulley系统与眼球,四条直肌及眼眶壁的三维立体模型的重建。同时应用3D-DOCTOR软件的测量工具,对Pulley及相关组织结构进行测量和分析。 结果 (1) Pulley位于眼球赤道部直肌穿过Tenon囊的部位,为纤维环状结构,包绕每条直肌。各直肌Pulley之间形成连接带。Pulley及其连接带的主要成分为胶原纤维、弹力纤维及平滑肌。内、外直肌Pulley通过与其成分相似的悬吊韧带与眼眶壁相连。各直肌肌肉纤维由眶层和球层组成,球层肌肉纤维穿过
[Abstract]:PurposeHistochemical and immunohistochemical techniques, continuous coronal section of the whole orbit and computer aided three-dimensional reconstruction were used to study the tissue composition and three-dimensional spatial distribution of human rectus muscle (Pulley) in the orbit.In order to provide anatomical and histological basis for exploring the function of Pulley, the morphologic structure of Pulley was demonstrated from different angles.Method1) 5 cases of fresh cadavers without ocular diseases were fixed and embedded with coronal sections of the whole orbit.After Masson and Weigert special staining and immunohistochemical staining, the tissue structure of each layer was observed from the general angle and under light microscope.(2) the typical tissue sections of Pulley and Pulley junctional bands were stained with picric acid-Sirius red to observe the composition and distribution of collagen in the Pulley and Pulley junctions of rectus muscle under polarized light microscope.The collagen composition of Pulley and Pulley junctions of four rectus muscles were analyzed by Mias-2000 image analysis system.3) the whole orbital coronal continuous tissue slice image stained by Masson was recorded by digital camera and then input into the computer one by one. The image was segmented with Pulley system and eyeball by photoshop 7.0 software to remove noise, and 3D-Doctor software was used to segment the image.Reconstruction of three-dimensional models of four rectus muscles and orbital walls.At the same time, the measurement tool of 3D-DOCTOR software is used to measure and analyze the Pulley and related organization structure.ResultPulley is located in the equatorial rectus of the eye through the Tenon sac and is a fibrous annular structure that encircles each rectus muscle.The main components of the connective band. Pulley and its connective band are collagen fiber, elastic fiber and smooth muscle.The medial and outer rectus Pulley are connected to the orbital wall through a suspension ligament similar to its composition.The rectus muscle fibers are composed of the orbital layer and the bulbar layer, and the bulbar muscle fibers pass through the orbital layer.
