【摘要】:背景: 据调查发现,,在过去五十年中男性精子质量呈下降趋势,少精症、无精症患者屡见不鲜,环境因素被认为是致病的主要因素,其中温度作为一种外在因素引起男性不育愈来愈被重视,研究表明:包括人类在内的大部分哺乳动物的睾丸需要一个比腹腔温度低的环境,睾丸暴露在腹腔温度或高于腹腔温度可导致一种可复性的生精障碍和生精细胞退化,可引起暂时的、部分或完全的不育。这种高温作用在不同的生长发育阶段,对其产生的影响可能是不同的。青春期是男性发育过程中第二个高峰期,在此期男性会出现第二性征并且生殖器官渐趋成熟,青春期对男性不仅在身体发育方面还是在性腺发育方面都具有重要意义,如此期受到外界环境不良干扰后可能会影响男性生殖功能和个体的生长发育。现今,男性不育患者日益增多,往往都是成年后才会发现,从而寻找原因给予相应的治疗,但一部分不育患者可能是在婴幼年或青少年期受到某种不良刺激后而引起的。目前随着医疗水平的提高,男性医学保健教育的广泛宣传,男性对自己的生殖健康提高重视,出现男性不育症愈来愈年轻化的现象。另外,越来越多的研究报道男性不育和氧化损伤有着密切的关系,活性氧是导致男性不育的罪魁祸首。因此,本研究试图探讨青春期前后大鼠睾丸局部高温对青春期前和青春期生精细胞凋亡的影响,同时并探讨生精细胞发生凋亡、氧化应激、睾酮三者之间的关系。 目的: 1) 研究青春期前后大鼠睾丸局部热作用后睾丸生精上皮的变化; 2) 探索睾丸局部热作用后睾丸组织氧化应激作用及其与生精细胞凋亡的关系; 3) 研究睾丸局部热作用后睾酮的变化及其与氧化应激的关系。 材料与方法: 雄性Sprague-Dawley(SD)青春期前大鼠(5周龄),分笼饲养于标准动物房(22℃),给予规律光照(12h∶12h),自由饮食,随机分热处理组和对照组,热处理组按热处理后2h、
[Abstract]:Background:According to the survey, in the past 50 years, the quality of male sperm has been declining, oligozoospermia and azoospermia are common, and environmental factors are considered to be the main cause of the disease.Temperature, as an external factor, has attracted increasing attention to male infertility. Studies have shown that the testicles of most mammals, including humans, need an environment with lower temperatures than that of the abdominal cavity.Testicular exposure to or above celiac temperature may lead to a refolding of spermatogenic disorders and degeneration of spermatogenic cells, which may cause temporary, partial or complete infertility.The effect of this high temperature on it may be different at different stages of growth and development.Adolescence is the second peak of male development, in which men develop secondary sexual symptoms and mature reproductive organs.
