雪旺细胞复合脱细胞细胞外基质预构建修复长段神经缺损的实验研究

【摘要】:背景 虽然桥接法修复长段神经缺损的标准做法是神经移植,但自体神经移植可供桥接长段神经缺损的供区有限,是一种创伤修复创伤,限制了临床应用。为了克服这一缺点,一直在继续研究寻找更易接受的替代物。目前的研究认为神经缺损的替代材料有:非生物材料PLA、PGA、PPE;生物材料:血管、膜管、变性肌肉、AECM;自体神经移植:游离移植、带血管蒂移植;组织工程:人工神经。AECM抗原性低,且具备接近人体的网架结构、良好的生物相容性、生物力学性能;FN,LN能较完整保留在细胞外基质内,其表面具备细胞膜受体识别位点,有助于促进与雪旺细胞黏附和神经轴突生长,因而是组织工程化人工神经的理想支架。但应用AECM修复缺损的距离受限制,组织工程学研究为解决周围神经修复难题带来新的希望。 目的 本研究采用组织工程化技术制备的脱细胞细胞外基质并行端侧吻合法预构建桥接物修复长段神经缺损,并且雪旺细胞复合脱细胞细胞外基质并行端侧吻合法预构建桥接物修复长段神经缺损,对桥接长段神经缺损的效果做了初步探索,旨在开发自体神经移植的理想替代材料。为组织工程人工神经的临床应用提供理论依据。 方法 第一部分:组织工程化神经桥接物制备(化学萃取法);脱细胞细胞外
[Abstract]:BackgroundAlthough the standard method for repairing long segment nerve defect by bridging method is nerve transplantation, the donor area of autogenous nerve graft for bridging long segment nerve defect is limited, which is a kind of trauma repair trauma, which limits the clinical application.To overcome this shortcoming, research has continued to find more acceptable alternatives.Current studies suggest that alternative materials for neural defects are: non-biomaterial PLA PGAPPE; biomaterials: blood vessels, membrane tubes, denatured muscles AECM; autologous nerve transplantation: free transplantation with vascular pedicle; tissue engineering: artificial nerve. AECM has low antigenicity.In addition, it has close to human network structure, good biocompatibility, and its biomechanical properties can be completely preserved in extracellular matrix, and its surface has cell membrane receptor recognition site.It is helpful to promote the adhesion of Schwann cells and the growth of nerve axons, so it is an ideal scaffold for tissue engineering artificial nerve.However, the distance of repairing defects with AECM is limited, and the study of tissue engineering brings new hope to solve the problem of peripheral nerve repair.PurposeIn this study, an acellular extracellular matrix (ECM) with end-to-side anastomosis was prepared by tissue engineering technique to repair long nerve defects.In addition, Schwann cells combined with acellular extracellular matrix and end-to-side anastomosis were constructed to repair long segment nerve defects.
