白念珠菌锌簇转录因子FCR1的功能研究

【摘要】:FCR1基因是白念珠菌锌簇转录因子,它能够负向调控白念珠菌的多药耐药现象,但其调控的分子机制目前尚不清楚。本课题利用药物敏感性实验、罗丹明6G外排实验、薄层色谱分析、RT-PCR、Realtime-PCR等方法研究白念珠菌锌簇转录因子FCR1参与多药耐药的分子机制,证明FCR1基因在氟康唑诱导的条件下能够负向调控多药耐药基因CDR1、CDR2和MDR1的表达,增强白念珠菌的药物外排功能,从而使白念珠菌产生抗药性。另外,基因芯片杂交技术发现FCR1缺失以后不同功能分类的基因表达差异,说明FCR1基因可能参与白念珠菌多种生命活动。其中菌丝和毒力相关基因NAG2和CTA1表达上调,RPL6基因表达下调,通过菌丝诱导和毒力实验证明FCR1缺失能够负向调控白念珠菌菌丝的生长和毒力。
[Abstract]:FCR1 gene is a zinc cluster transcription factor of Candida albicans, which can negatively regulate the multidrug resistance of Candida albicans, but its molecular mechanism is still unclear.In this study, drug sensitivity test, Rhodamine 6G efflux test and TLC analysis were used to study the molecular mechanism of zinc cluster transcription factor (FCR1) in Candida albicans involved in multidrug resistance.It is demonstrated that FCR1 gene can negatively regulate the expression of CDR1, CDR2 and MDR1 under fluconazole-induced condition, and enhance the drug efflux function of Candida albicans, thus causing Candida albicans to develop drug resistance.In addition, the gene expression of different functional classifications after FCR1 deletion was found by gene chip hybridization, indicating that FCR1 gene may be involved in many kinds of life activities of Candida albicans.Among them, hyphal and virulence related genes NAG2 and CTA1 up-regulated the expression of RPL6 gene. It was proved by hyphal induction and virulence experiment that FCR1 deficiency could negatively regulate the growth and virulence of Candida albicans.
