活化蛋白C对大鼠急性胃粘膜损伤保护治疗作用的实验研究

【摘要】:目的探讨活化蛋白C(APC)对急性胃粘膜损伤的保护治疗作用。 材料及方法大鼠48只,随机分为八组。A1、A2组为正常组,B1(WRS2h)、B2(WRS3.5h)组为模型组,C1(WRS2h)、C2(WRS3.5h)组为rhAPC治疗组,D1(WRS2h)、D2(WRS3.5h)组为生理盐水对照组。复制浸水束缚应激(WRS)模型并计算溃疡指数(UI)。C1、C2组在WRS开始前半小时通过鼠尾静脉注入rhAPC(100ug/kg),D1、D2组在同一时间点注入同等量的生理盐水。分离胃粘膜上皮细胞,并调制成细胞浓度为1×106个/ml的单细胞悬液,加入DiOC6,上流式细胞仪检测线粒体跨膜电位(Δψm)。采用ELISA法检测各组血清中TNF-α、IL-1β、IL-6的浓度。取胃底腺部分胃粘膜上皮组织,10%甲醛固定48小时后,经HE染色,光镜下观察。 结果在WRS各时间点UI,TNF-α、IL-6的浓度均较正常组明显升高(P0.01),在WRS2h后TNF-α(647.8±87.55pg/ml)、IL-6(52.85±5.51pg/ml)比WRS3.5h后TNF-α(474.3±103.6)、IL-6(35.6±5.72)为高;胃粘膜的病理损害以WRS3.5h后更明显。线粒体跨膜电位较正常组明显下降,WRS2h后为78.80±33.19,WRS3.5h后为52.83±20.89;IL-1β较正常组有下降趋势,但无显著差异(P0.05)。rhAPC治疗组各时间点TNF-α、IL-6的浓度均较模型组显著下降(P0.001), IL-1β的浓度较模型组显著升高(P0.01),线粒体跨膜电位较模型组显著升高(P0.01),胃粘膜的损伤得到减
[Abstract]:Objective to investigate the protective and therapeutic effects of activated protein (APC) on acute gastric mucosal injury.The model of water-immersion restraint stress (WRS) was established and the ulcer index was calculated. The ulcer index was calculated. Half an hour before the onset of WRS, the rats were injected with the same amount of normal saline through the tail vein of rhAPC-100ugr / kg ~ (-1) D _ (1) D _ (2) group at the same time.Gastric mucosal epithelial cells were isolated and single cell suspensions of 1 脳 106 / ml were modulated. DiOC6 was added. Mitochondrial transmembrane potential (螖 蠄 m) was measured by upflow cytometry.The concentration of TNF- 伪 and IL-1 尾 and IL-6 in serum of each group was detected by ELISA method.The gastric fundus gland was fixed with 10% formaldehyde for 48 hours, then stained with HE and observed under light microscope.Compared with the normal group, the mitochondrial transmembrane potential decreased significantly from 78.80 卤33.19 WRS3.5h to 52.83 卤20.89mil IL-1 尾 after 2h compared with the normal group.However, there was no significant difference in the concentration of TNF- 伪 and IL-6, the concentration of IL-1 尾 in the model group was significantly higher than that in the model group, the mitochondrial transmembrane potential was significantly higher than that in the model group, and the gastric mucosal damage was decreased.
