基于单核-巨噬细胞炎症模型的络风宁1号抗炎机制初探

【摘要】:目的制备单核巨噬细胞炎症模型,并观察络风宁1号含药血清对模型细胞增殖的影响。方法采用脂多糖(LPS)体外诱导建立单核巨噬细胞炎症模型,造模的同时加入不同浓度的络风宁1号及匹伐他汀钙含药血清干预,共同孵育24 h后,收集细胞上清,ELISA法检测炎症因子基质金属蛋白酶9(MMP 9)水平。结果与空白组相比,模型组THP 1单核细胞显著升高(P0.01);不同浓度络风宁1号含药血清干预后,高浓度组的抑制作用最明显(P0.01)。结论单核巨噬细胞炎症模型制备成功,基于此模型,络风宁1号含药血清可以抑制LPS对炎症细胞的增殖作用,这可能是其抗炎症、防治冠心病不稳定性心绞痛的主要机制之一。
[Abstract]:Objective to establish a mononuclear macrophage inflammatory model and to observe the effect of Luofuning 1 serum on the proliferation of the model cells.Methods the mononuclear macrophage inflammatory model was induced by lipopolysaccharide (LPS) in vitro. The model was incubated with different concentrations of Luofenin 1 and pivastatin calcium containing serum for 24 h.The level of inflammatory factor matrix metalloproteinase (9(MMP 9) was detected by Elisa.Results compared with the blank group, the monocyte of THP 1 in the model group was significantly higher than that in the control group, and the inhibitory effect of the high concentration group was the most obvious after the intervention of different concentrations of Luofunning 1 containing serum.Conclusion the mononuclear macrophage inflammatory model was successfully prepared. Based on this model, the drug serum Luofuning 1 can inhibit the proliferation of LPS on inflammatory cells, which may be one of the main mechanisms of anti-inflammation and preventing and treating unstable angina pectoris of coronary heart disease.
