ALF在人精子发生过程中的作用及对男性生育的影响

【摘要】:睾丸的精子发生是一个独特复杂的细胞分裂分化过程。睾丸生精小管中的生精细胞先后经过三个阶段的变化:精原细胞的增殖,精母细胞的减数分裂和精子细胞的变态过程,最终产生成熟的精子。许多在睾丸中特异或高度表达的基因参与这一过程。克隆睾丸精子发生相关基因并研究其功能,对于揭示精子发生过程中减数分裂和变态的分子机制,对于临床男性不育的诊断和治疗以及男性分子节育等均具有重要的意义。 本实验室先期利用人睾丸cDNA microarray杂交技术比较了人胚胎期睾丸和成年期睾丸的基因表达差异,筛选出386个差异表达基因,ALF是其中的一条睾丸特异表达的基因,它是通用转录因子TFⅡA大亚单位(TFⅡAα/β)的类似物,,是生殖细胞特异形式的转录因子。本研究着重于ALF在人精子发生中的作用,以及其表达水平的改变与男性生育之间的关系。cDNA芯片杂交结果显示ALF高表达于成人睾丸组织,在胚胎睾丸中表达量较低;16例生精障碍患者睾丸的RT-PCR结果显示:ALF在2例唯支持细胞综合症病人睾丸中表达缺失;在7例生精阻滞患者睾丸中,有一例没有表达,另外6例有强弱不等的表达;在7例生精紊乱患者睾丸中,1例表达缺失。原核表达ALF重组蛋白,纯化并免疫小鼠制备多克隆抗体,经Western Blot分析显示ALF抗体在人睾丸和精子中均显示有特异条带,其分
[Abstract]:Testicular spermatogenesis is a unique and complex process of cell division and differentiation. Spermatogenic cells in testicular seminiferous tubules undergo three stages: spermatogonia proliferation, spermatocyte meiosis and sperm cell metamorphosis, resulting in mature spermatozoa. Many genes that are specific or highly expressed in the testis are involved in this process. Cloning testicular spermatogenesis related genes and studying their functions are of great significance in revealing the molecular mechanism of meiosis and metamorphosis during spermatogenesis, in the diagnosis and treatment of clinical male infertility and in male molecular sterility. The difference of gene expression between human embryonic testis and adult testis was compared by using human testis cDNA microarray hybridization technique, and 386 differentially expressed genes were screened out.
