幽门螺杆菌融合蛋白UreB-Omp11在大肠杆菌中的表达与鉴定

【摘要】:幽门螺杆菌(Helicobacter pylori,Hp)感染是世界上最常见的感染之一,目前Hp已经成为公认的人类慢性活动性胃炎和消化性溃疡的主要致病因子,并与胃癌、胃MALT恶性淋巴瘤的发生密切相关。目前,临床上主要采用抗生素治疗Hp的感染,短期内虽有较好的疗效,但由于存在细菌耐药性不断增加、治疗费用高、不能防止复发和重复感染等问题,使这类方案的实用性受限。疫苗接种经济而简便,可在人群中大规模运用,并且对耐药细菌仍然有效,因此研究幽门螺杆菌疫苗有重要意义。 虽然Hp疫苗研究取得了一定成绩,但一直令学者们遗憾的是单一抗原成分免疫动物时,获得的保护率均不理想,,为了克服单种抗原免疫性较弱的不足有学者建议将多种抗原成分联合起来制作疫苗。这就为今后Hp疫苗的探索和研究指出了发展方向。 国内外大量研究证实UreB有较好的免疫保护效果,而且已有研究将UreB疫苗制剂进入到二期临床试验,证实Hp尿素酶在人体有效。已有研究结果显示Hp外膜蛋白中有些具有良好的免疫学活性,Omp11是Hp的一种外膜蛋白,其编码基因具有高度保守性,经动物实验鉴定亦取得了较好的免疫保护效果,为重要的免疫抗原。
[Abstract]:Helicobacter pylori (Helicobacter pylori) infection is one of the most common infections in the world. At present, HP has become the main pathogenic factor of chronic active gastritis and peptic ulcer in human, and is closely related to the occurrence of gastric cancer and gastric MALT malignant lymphoma. At present, antibiotics are mainly used in the treatment of HP infection in the short term, but because of the increasing drug resistance of bacteria and the high cost of treatment, it can not prevent recurrence and repeated infection. The practicability of such schemes is limited. Vaccination is economical and simple, can be used in large scale in population, and is still effective against drug-resistant bacteria, so it is of great significance to study Helicobacter pylori vaccine. Although some achievements have been made in the research of HP vaccine, it has always been a pity to scholars that the protective rate obtained when immunizing animals with a single antigen component is not satisfactory. In order to overcome the weak immunogenicity of a single antigen, some scholars have suggested that a variety of antigen components should be combined to make a vaccine. This pointed out the direction of development for the exploration and research of HP vaccine in the future. A large number of studies at home and abroad have confirmed that UreB has a good immune protection effect. Moreover, it has been studied that UreB vaccine preparation has been put into the second phase clinical trial, which proves that HP urease is effective in human body. It has been shown that some of HP outer membrane proteins have good immunological activity. Omp11 is an outer membrane protein of HP, and its coding gene is highly conserved. Is an important immune antigen.
