两种宿主来源毛首线虫形态学与分子生物学的分类研究

【摘要】:本研究从重庆市动物园阿拉伯狒狒和松鼠猴体内收集了两种毛首线虫成虫。为了初步鉴定虫种并探讨这两种毛首线虫与已知六种毛首线虫的亲缘距离,我们对这两种毛首线虫进行了形态学鉴别和分子生物学方法的分类学研究。 本研究利用测微尺对虫体体长、前部长、后部长、输精管长、射精管长、泄殖腔长、交合刺长、交合刺鞘长,交合刺鞘刺高度等(测量单位为 mm)形态学体征进行了测量并显微镜拍照。根据所获得的测量数据和显微镜观察雄虫生殖器官形态学特点,与已知虫种进行比对,做出形态学初步鉴别。随后,抽提两种毛首线虫的 DNA,通过 PCR 扩增出核糖体 DNA(rDNA)中的 ITS2 片断,进行测序,并使用 DNAMAN等软件与已公布的六种毛首线虫的 ITS2 数据比较亲缘距离等数据,进行分类学研究。最后,将 PCR 产物纯化,选用 MspⅠ和 HaeⅢ两种限制性内切酶对纯化产物进行 RFLP 分析,并获得酶切图谱。 结果显示,这两种不同来源的毛首线虫在形态学上有明显区别,其中阿拉伯狒狒来源的毛首线虫测量数据在 T. cynocephalus(皮翼毛首线虫)参考范围内,雄虫生殖器官外形特征与 T. cynocephalus(皮翼毛首线虫)相近,初步鉴定为 T. cynocephalus(皮翼毛首线虫)。松鼠猴来源毛首线虫的测量数据和对雄虫生殖器官的观察结果表明这种毛首线虫与 T. rhinopiptheroxell-a sp.nov(川金丝猴新种毛首线虫)相似,由于获得松鼠猴来源毛首线虫成虫少于形态学鉴别所需成虫数的最小值(≥10 条),所以无法确定,疑为 T. rhinopiptheroxell-a sp.nov(川金丝
[Abstract]:Two species of adult nematodes were collected from Arabian baboon and squirrel monkey in Chongqing Zoo. In order to identify the two species and to study the relationship between the two species and six known species of nematode, we studied the morphological identification and molecular biological methods of the two species of nematode. In this study, the length of the body, the former minister, the posterior head, the vas deferens, the ejaculatory duct, the cloaca, the intersecting spines and the sheaths of the intersecting spines were measured by micrometer. The morphologic signs, such as height of spiny spines (measured in mm), were measured and photographed under microscope. According to the measured data and microscopic observation, the morphological characteristics of male reproductive organs were compared with those of known species, and the morphological characteristics were preliminarily identified.
