骨髓基质干细胞复合几丁质凝胶体外联合培养的实验研究

【摘要】:前言 可注射性水凝胶是国际上近几年兴起的一种相对新颖的软骨组织工程材料,它提供了一类新的细胞固定机制,可注射性材料用于软骨缺损修复的优势在于材料与修复细胞之间嵌合连接好、塑形容易,细胞和生长因子能与材料充分均匀混合,且能通过关节镜或其它微创方式操作,安全、有效、费用低。本实验利用可注射性水凝胶——几丁质作为骨髓基质干细胞载体,在体外进行联合培养,为构建组织工程软骨组织提供实验依据。 材料与方法 1 实验动物 成年SD大鼠10只,雄鼠体重120—140g,由中国医科大学实验动物中心提供。 2 主要试剂和仪器 几丁质(SIGMA公司),DMEM(GIBCO公司产品),新生牛血清(GIBCO公司产品),percoll梯度分离液(PHARMACIA公司产品密度:1.073g/m),胰岛素样生长因子IGF-Ⅰ、转化生长因子TGF-β_1(Sigma公司)、维生素C、地塞米松,CO_2培养箱,倒置显微镜。 3 几丁质凝胶的制备 在10℃下,将几丁质(SIGMA公司)10g放入研钵中,加丙酮40ml研磨10min;边研磨边缓缓加入冷浓HC1400ml,充分研磨,使呈糊状。于4℃放置24h,之后用玻璃棉过滤,将滤液缓缓加入到盛有2000ml体积分数50%乙醇的烧杯中,边加边剧烈搅拌,然后静置,待胶态几丁质析出后去除清液,收
