Tenascin与大鼠胚胎心脏发育关系的研究

【摘要】: 目的:检测韧黏素-C(TN-C)和韧黏素-X(TN-X)在不同孕龄大鼠胚胎心脏中的表达规律,探讨其在大鼠胚胎心脏发育过程中的作用。 方法:60只雌性SD大鼠,24只雄性SD大鼠,雌雄同笼,以观察到雌鼠排出阴栓为妊娠0.5天(ED0.5),分别于ED11.5、ED12.5、ED13.5、ED15.5、ED17.5、ED19.5、新生1天处死孕鼠或新生鼠,取出心脏或全胚。用半定量逆转录聚合酶链反应(RT-PCR)测定TN-C和TN-X在各孕龄的胚胎心脏中mRNA的表达水平;免疫组织化学染色法检测上述各孕龄大鼠心脏TN-C和TN-X蛋白的表达;免疫印迹法(Western blot)检测TN-C在大鼠胚胎心脏中的表达水平,进一步验证免疫组化的结果。 结果:TN-C mRNA和蛋白在大鼠胚胎心脏发育早期有丰富表达,于ED15.5即显著下调;TN-X mRNA在大鼠胚胎ED12.5开始表达上调,其后各孕龄表达未见明显差异。免疫组化显示,TN-C在ED11.5、ED12.5、ED13.5表达于心脏流出道、前心外膜器官、房室垫和心包腔,而在ED15.5、ED17.5、ED19.5、新生1天相应表达为阴性;TN-X在各孕龄胚胎心脏中均有表达,且随着胎龄增大,表达范围更广泛,于新生1天的大鼠心脏中也广泛表达。 结论:TN-C在大鼠胚胎心脏发育早期表达丰富,对心脏的早期发育有一定关系;而TN-X在ED11.5于形成血管的心外膜细胞周围表达,提示TN-X可能与心脏早期冠脉等血管的形成有关。
[Abstract]:Aim: to detect the expression of tensionin-Con-TN-C) and ductile-XTN-X) in rat embryonic heart of different gestational ages and to explore the role of them in the development of rat embryonic heart. Methods Twenty four male Sprague-Dawley rats with 60 female Sprague-Dawley rats were treated in the same cage. It was observed that the female rats excreted negative suppository as 0.5 day gestation ED0.5, ED11.5 ED12.5 ED15.5ED15.5ED17.5ED19.5. on the first day of birth, pregnant rats or newborn rats were killed and the heart or whole embryo was taken out. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of TN-C and TN-X in the fetal heart of each gestational age, and the expression of TN-C and TN-X protein in the heart of each gestational age was detected by immunohistochemical staining. Western blots were used to detect the expression of TN-C in rat embryonic heart. Results there were abundant expression of TN-C mRNA and protein in the early stage of embryonic heart development in rats. After ED15.5, the expression of TN-X mRNA began to up-regulate in rat embryo ED12.5, but there was no significant difference in the expression of TN-C mRNA and protein in different gestational ages. Immunohistochemical staining showed that TN-C was expressed in cardiac outflow tract, anterior epicardial organ, atrioventricular pad and pericardium, and ED19.5at ED15.5ED17.5. at the first day of birth, TN-X was expressed in the heart of every gestational age, and it increased with gestational age. The expression was more widespread and was also widely expressed in the heart of a day-old rat. Conclusion the expression of TN-C is abundant in the early stage of embryonic heart development in rats, which may be related to the early development of the heart, while TN-X is expressed around the epicardial cells that form blood vessels, suggesting that TN-X may be related to the formation of coronary arteries and other vessels in the early stage of the heart.
