卵子成熟机理的蛋白质组学研究

【摘要】:随着社会发展,不孕症越来越成为困扰夫妻双方身心健康的世界性问题。世界卫生组织预测,在21世纪,不孕症将成为仅次于肿瘤和心脑血管病的第三大疾病。故不孕不育症是一项世界性医学和社会学的重要问题,也是当前生命科学研究的热点。随着辅助生育技术的日益发展,越来越多的不孕症患者由助孕的方法获得妊娠。在对辅助生殖技术的研究过程中,人们深刻认识到了如何得到和保留优质卵子的重要性。卵母细胞发生、发育、成熟的调节机制的一直是研究的焦点。近年来,人们对卵母细胞和卵泡细胞发育的分子机制进行了大量的研究,但大多采用单通道模式,即对单个蛋白、基因的功能研究。蛋白质是基因功能的实施者,蛋白质有自身特有的活动规律,如动态修饰、加工、转运定位、结构形成、代谢等,均无法从基因组水平上的研究获知。而且卵子发育过程是一个涉及众多信号分子的调控网络,包括许多已知及未知的重要蛋白参与,采用单通道的模式无法进行高效的研究。从蛋白质水平对卵子发育机制进行高通量的研究是本课题的目标。 20世纪90年代中期,国际上萌发了蛋白质组学这一新的学术领
[Abstract]:With the development of society, infertility has become a world-wide problem that puzzles the physical and mental health of husband and wife. The World Health Organization predicts that infertility will become the third largest disease in the 21st century after cancer and cardiovascular and cerebrovascular diseases. So infertility is an important problem in medicine and sociology all over the world, and it is also a hot topic in life science research. With the development of assisted fertility technology, more and more infertility patients get pregnancy by the aid of pregnancy. In the course of research on assisted reproductive technology, people have realized the importance of how to obtain and retain high quality eggs. The regulation of oocyte formation, development and maturation has been the focus of research. In recent years, a great deal of research has been done on the molecular mechanism of oocyte and follicle cell development, but most of them adopt single channel mode, that is, the function of single protein and gene. Proteins are the implementers of gene function. Proteins have their own specific activity rules, such as dynamic modification, processing, transport location, structure formation, metabolism and so on. Furthermore, egg development is a regulatory network involving many signaling molecules, including many known and unknown important proteins, which can not be efficiently studied using a single channel model. High-throughput research on egg development mechanism from protein level is the goal of this paper. In the mid-1990s, proteomics, a new academic field, came into being in the world.
