【摘要】:腹侧盖膜区(ventral tegmental area,VTA)的多巴胺细胞与回报行为以及认知功能有着密切的关系。多巴胺细胞有两种放电模式,规律放电和 bursting。Bursting 这种放电模式与回报行为有关,而在细胞学上其可以增加突触内神经递质多巴胺的含量。在大鼠的研究中,apamin 和 NMDA 都可以在多巴胺细胞上诱发 bursting,但其作用机理不同,apamin 诱发的 bursting 可以被 nifedipine 抑制,而相反 NMDA诱发的 bursing 不能被 nifedipine 抑制。这提示在生物系统中,bursting这种放电模式可以通过不同的两套系统实现。但是 apamin 诱发的bursting 是依赖哪种神经传导系统尚没有定论。而目前的研究大多数集中在大鼠,对小鼠放电行为的研究很少。故本课题着重探讨了小鼠VTA 区多巴胺细胞放电的行为模式,同时试图探讨 NMDA 和carbachol 诱发的 bursting 的不同机理。 1. 体外小鼠 VTA 区多巴胺细胞主要存在两种放电模式。 研究结果指出:在室温 perforated 膜片钳技术下,小鼠 VTA 区多巴胺细胞 31%静止不自发放电,65%呈现规律或者不规律的放电模式,4%呈现自发 bursting 的放电模式。 2. NMDA 诱发的 bursting 不是由钙离子介导的。 研究结果指出:NMDA 可以在 32%记录的小鼠 VTA 区的多巴胺细胞上诱发出 bursting,其不能被电压依赖性钙离子通道的抑制剂抑制(n=5);同时用 TTX 抑制 sodium spikes 后呈现出的膜电位震荡同样不能被电压依赖性钙离子通道的抑制剂抑制(n=3)。
[Abstract]:The dopamine cells of ventral tegmental are closely related to reward behavior and cognitive function. There are two discharge modes in dopamine cells: regular discharge and bursting.Bursting, which are related to the behavior of reward, but it can increase the content of dopamine in synaptic neurotransmitter in cytology. In the study of rats, both apamin and NMDA could induce burstling in dopamine cells, but bursting induced by apamin in different mechanisms could be inhibited by nifedipine, but bursing induced by NMDA could not be inhibited by nifedipine. This suggests that in biological systems bursting can be implemented in two different systems. But it is not clear what kind of nerve conduction system is dependent on bursting induced by apamin. However, most of the current studies are focused on rats, and little research has been done on the discharges in mice. Therefore, this study focused on the behavioral pattern of dopamine cell discharges in VTA region of mice, and tried to explore the different mechanisms of bursting induced by NMDA and carbachol. 1. There are two kinds of discharge patterns in dopamine cells in VTA region of mice in vitro. The results showed that at room temperature perforated patch clamp technique, 31% of the dopamine cells in the VTA region of mice showed regular or irregular discharge patterns (65%), or 4% (4%) of the dopamine cells in the VTA region showed spontaneous bursting discharge patterns. 2. Bursting induced by NMDA is not mediated by calcium ion. The results showed that the burstingwas induced in 32% of the mouse VTA dopamine cells, which could not be inhibited by the voltage-dependent calcium channel inhibitor, and the membrane electricity after sodium spikes was inhibited by TTX. Position oscillation also cannot be inhibited by voltage-dependent calcium channel inhibitors.
