【摘要】:流感病毒是最常见的呼吸道致病因子,每次流感大流行的时候都有很多人死亡,例如在1918年的流感大流行就有2000万人死于流感。流感病毒常见的宿主有人、猪、马和禽类,并可以在不同宿主之间交叉传播,如1997年香港的流感病毒就是由鸡传播给人的,在18个感染流感的人中就有6个死亡。疫苗是预防流感的一种好方法,目前最常用的是灭活疫苗(Ivac),其次是减毒活疫苗(Livac)和亚单位疫苗(Subvac)。流感DNA疫苗是一种新型的预防流感的疫苗,目前正在很多国家的实验室中进行研究,部分已经在动物实验中获得了良好效果。 DNA疫苗能够同时刺激机体B细胞,辅助性T细胞和细胞毒性T细胞,产生特异性体液免疫和细胞免疫,因而受到大家关注。美国国立卫生研究院(NIH)和世界卫生组织(WHO)都强调流感DNA疫苗是流感疫苗研究的一个重要方向,流感病毒血凝素(hemagglutinin,HA)基因的研究正是该重要研究内容之一。流感病毒基因组第4节段编码HA,人和动物流感病毒的HA刺激机体产生最主要的保护性抗体,此抗体具有中和病毒和阻止病毒穿入靶细胞的作用,能中断病毒的复制。HA分为重链HA1和轻链HA2区,HA1区是该蛋白与宿主细胞受体结合部分,HA2区是具有膜融合活性的部分。本文对流感病毒HA1区进
[Abstract]:Influenza viruses are the most common respiratory factor, with many people dying during each pandemic, such as the 1918 pandemic, which killed 20 million people. Influenza virus is a common host of people, pigs, horses and birds, and can be spread across different hosts. For example, in 1997, the influenza virus in Hong Kong was transmitted from chickens to humans, and six of the 18 people infected with the flu died. Vaccine is a good way to prevent influenza. At present, the most commonly used vaccine is inactivated vaccine Ivaca, followed by live attenuated vaccine (livaca) and subunit vaccine. Influenza DNA vaccine is a new vaccine to prevent influenza. It is being studied in many laboratories in many countries, and some of it has achieved good results in animal experiments. DNA vaccine can stimulate B cells, helper T cells and cytotoxic T cells at the same time and produce specific humoral and cellular immunity. Both the National Institutes of Health (NIH) and the World Health Organization (WHO) have emphasized that influenza DNA vaccine is an important direction of influenza vaccine research, and the study of influenza virus hemagglutinin (HA) gene is one of the important research contents. Section 4 of the influenza virus genome encodes HA.The HA of human and animal influenza viruses stimulates the body to produce the most important protective antibody, which neutralizes the virus and prevents it from penetrating into the target cells.
