P物质对大鼠睡眠—觉醒周期和情绪活动的影响及其作用机制

【摘要】:目的:应用情绪行为活动测定、脑立体定位、侧脑室插管、微量注射、多导睡眠描记和免疫组织化学等方法,观察P物质和其拮抗剂(N-acetyl-L-tryptophan 3,5-bis(trifluoromethyl)-benzyl ester,NALTBE)对大鼠睡眠-觉醒周期及其情绪活动的影响。 方法:80只成年雄性Sprague-Dawley大鼠随机分为睡眠记录组和情绪行为组。睡眠记录组大鼠经水合氯醛(350mg/kg)腹腔注射麻醉后,将头部固定于SN-2型脑立体定位仪上,暴露颅骨,双氧水清洁颅骨表面,在冠状缝前2mm及人字缝前1mm与颅骨中线两侧旁开1mm交叉点处分别安装EEG电极,,深度以穿透颅骨接触到硬脑膜为度,用于记录皮层脑电活动;在双侧颈肌内插入EMG电极用于记录肌电活动。EEG、EMG记录电极引导线均连接到一个微型插座上。外径为23gauge(0.6mm,OD)的不锈钢引导管按Paxinos和Watson大鼠脑立体定位图谱插入:右侧脑室(AP:-0.8mm,R:+1.5mm、H:-4mm),引导管顶端距侧脑室1mm,供侧脑室内微量注射药物用。所有电极和引导管均用牙科水泥固定于大鼠的颅骨上。情绪行为组大鼠只埋入引导管,不安装电极。 手术后,大鼠在记录舱内饲养1周以恢复和适应环境。睡眠记录组进一步分为:PBS注射组(PBS,0.1M,PH=7.4,n=8);SP 1 pmol组(n=8);SP 10 pmol组(n=8);SP 100 pmol组(n=8);Anti-sP组(N-acetyl-L-tryptophan
[Abstract]:Objective: to investigate the effects of mood and behavioral activity determination, stereotactic localization, lateral ventricular intubation, microinjection, polysomnography and immunohistochemistry. To investigate the effects of substance P and its antagonist N-acetyl-L-tryptophan (N-acetyl-L-tryptophan) on sleep-wake cycle and emotional activity in rats. Methods 80 adult male Sprague-Dawley rats were randomly divided into sleep recording group and emotional behavior group. Rats in the sleep recording group were anesthetized by intraperitoneal injection of chloral hydrate (350mg / kg). The head was fixed on the SN-2 brain stereotactic locator to expose the skull, and hydrogen peroxide was used to clean the surface of the skull. EEG electrodes were installed at 2mm before coronal suture and 1mm crossing point between 1mm and midline of skull before herringbone suture. The depth of EEG electrode was used to record cortical electroencephalogram (EEG) activity by penetrating the skull into the dura mater. The EMG electrode was inserted into the bilateral cervical muscles to record the electromyoelectric activity. The EEGG EMG recording electrode guide wire was connected to a micro socket. The stainless steel guide tube with external diameter of 23gauge.6mm OD) was inserted according to the stereotaxic map of Paxinos and Watson rats. The right ventricle (AP: 0.8mm) was inserted as follows: the right ventricle (AP: 0.8mm), the right ventricle (1.5 mm), the apical lateral ventricle (1mm), and the donor ventricle for microinjection. All electrodes and guide tubes were fixed to the skull of rats with dental cement. In the emotional behavior group, only the guide tube was embedded, and the electrodes were not installed. After operation, rats were kept in the recording chamber for 1 week to recover and adapt to the environment. The sleep record group was further divided into two groups: PBS0.1 pmol 7.4MnPBS, SP1 pmol, SP10 pmol, SP100 pmol, N-acetyl-L-tryptophan (N-acetyl-L-tryptophan).
