脂肪来源基质细胞的分离扩增及成脂肪诱导分化研究

【摘要】: 脂肪来源的基质细胞(adipose derived stromal cells, ADSCs)是近年来从脂肪抽吸物中发现的一类成体干细胞,与较早被认识的成体干细胞骨髓基质细胞(bone marrow stromal cells, BMSCs)有很多相似的特性,并且脂肪组织来源丰富,脂肪抽吸术简单易行,自体移植时无免疫排斥等诸多优点。 本研究建立了人脂肪脂肪来源基质细胞(hADSCs)的分离和扩增方法,探讨ADSCs向脂肪细胞方向分化的潜能。采用胶原酶消化法分离扩增人ADSCs,测定其生长的经时变化、累积生长倍数等;应用不同因子诱导ADSCs向成脂细胞分化并进行鉴定。结果表明,每100ml脂肪抽吸物中可分离得到2×107~4×107个有核细胞,60d内扩增至第8代;油红O染色、免疫细胞化学染色及RT-PCR证实ADSCs经诱导可分化为脂肪细胞。ADSCs有望作为一种成体多能干细胞应用于组织工程及细胞治疗。
[Abstract]:Adipose derived stromal cells, ADSCs) is a kind of adult stem cells found in adipose aspirates in recent years. It has many similar characteristics with bone marrow stromal cells, BMSCs), a bone marrow stromal cell of adult stem cells known earlier. There are many advantages such as abundant adipose tissue sources, simple and easy fat aspiration, no immune rejection in autologous transplantation and so on. In this study, we established a method for the isolation and amplification of human adipose derived stromal cells (hADSCs), and explored the potential of ADSCs to differentiate into adipocytes. Human ADSCs were isolated and amplified by collagenase digestion, their growth time changes and cumulative growth times were measured, and different factors were used to induce ADSCs to differentiate into adipogenic cells and identify them. The results showed that 2 脳 10 ~ 7 ~ 4 脳 10 ~ 7 nucleated cells could be isolated from each 100ml fat aspirate and amplified to the 8th passage within 60 days. Immunocytochemical staining and RT-PCR confirmed that ADSCs could differentiate into adipocytes. ADSCs could be used as a kind of adult pluripotent stem cells in tissue engineering and cell therapy.
