抗甲3型流感病毒卵黄抗体的研究

【摘要】: 流行性感冒是由流感病毒(Influenza virus)引起的急性呼吸道传染病,由于流感病毒易变异的特性,新型流感病毒的出现常引起该病不同程度的流行。是我国当前流行最为广泛、危害性最严重的一种传染病。 从本世纪中叶开始,人们发现鸟类胚胎及新生儿能从母体获得大量的IgG抗体,鸡蛋蛋黄中也含有大量来源于血清的免疫蛋白IgG,且含量远高于血清,被命名为鸡卵黄免疫蛋白(Egg yolk immunoglobulin,IgY)。本实验采用H3N2型流感病毒通过两种途径免疫母鸡,以水稀释-硫酸铵沉淀及离子交换层析法提纯,并进行理化性质分析和动物效力试验型制备出高效价的抗甲3型流感病毒IgY.卵黄抗体制备方法简单,成本低,使用方便,且可广泛应用于人类及禽类疾病的预防,诊断和治疗,具有广阔的市场开发和综合利用的前景。
[Abstract]:Influenza is an acute respiratory infectious disease caused by influenza virus (Influenza virus). Is our country at present the most widespread, the harm most serious kind of infectious disease. Since the middle of this century, it has been found that birds embryos and newborns can obtain a large amount of IgG antibodies from their mothers, and egg yolks also contain a large number of immunoglobulin (IgG) from serum, which is much higher than that of serum. It was named as Egg yolk immunoglobulins (IGY) of chicken egg yolk. In this experiment, hens were immunized with H3N2 influenza virus through two ways, purified by water dilution ammonium sulfate precipitation and ion exchange chromatography, and the high titer anti-influenza virus IgY was prepared by physicochemical properties analysis and animal potency test. The preparation method of yolk antibody is simple, low cost, easy to use, and can be widely used in the prevention, diagnosis and treatment of human and poultry diseases. It has broad market development and comprehensive utilization prospects.
