人子宫内膜对胚胎接受性的初步研究

【摘要】: 人类子宫内膜对胚胎的接受性存在时间和空间的短暂性,即胚胎着床活动受到“种植窗”的限制,“种植窗”的启动常常发生在内源性黄体生成素(luteinizing hormone, LH)峰或使用外源性人绒毛促性腺激素(human chorionic gonadotropin, hCG)(LH的类似物)或黄体酮的第4-5天,关闭发生在第9-10天。进入到宫腔的胚胎和子宫内膜按一定的时、空顺序分泌相关蛋白或/和局部因子从而达到相互识别、相互融合完成着床过程。胚胎着床过程的限速步骤是胚胎发育与子宫内膜向接受态转化过程同步。而LH峰是促进卵子成熟、胚胎发育和子宫内膜向接受态同步转化的、最关键的调节因素。 如何评估胚胎发育与子宫内膜向接受态同步转化是预测胚胎着床成功与否的重要指标。 现行评估胚胎发育潜能的方法包括胚胎形态学评分和生化指标检测。然而胚胎评分与胚胎发育潜能并不完全正相关;胚胎分泌的细胞因子和蛋白类物质参与自身发育调节和胚胎着床过程,被预测可能用来衡量胚胎发育潜能的标志物。本论文将在我们前期工作基础上进一步进行人类卵母细胞和种植前胚胎LH表达的研究,为探讨LH在人类卵子和胚胎发育过程中的调节作用提供实验依据。上个世纪五十年代开始,学者们就从组织形态学、亚细胞结构、局部因子表达及其类固醇激素的调节作用等层次对子宫内膜胚胎接受性的评价方面做了大量研究,提出上皮细胞吞饮泡(pinopod)、整合素αυβ3、白血病抑制因子(LIF)、血管内皮生长因子(VEGF)的表达部分地反映子宫内膜对胚胎的接受性。但由
[Abstract]:The temporal and spatial transience of human endometrial receptivity to embryos, that is, the implantation of embryos is limited by the "implantation window". The initiation of "implant window" often occurs on the 4-5 day of endogenous luteinizing hormone (LHK) peak or the use of exogenous human chorionic gonadotropin (hCG)(LH analogue) or progesterone, and the closure occurs on the 9th to 10th day. When the embryo and endometrium enter the uterine cavity according to a certain order the empty sequence secretes the related protein or / and local factor to achieve mutual recognition and fusion to complete the implantation process. The limiting step of embryo implantation is the synchronization of embryo development and endometrial to receptive transformation. LH peak is the most important regulatory factor to promote the ovum maturation, embryo development and endometrial synchronous transformation to the receptive state. How to evaluate the synchronous transformation between embryo development and endometrium is an important index to predict the success of embryo implantation. Current methods for evaluating embryonic development potential include embryo morphological scores and biochemical indicators. However, embryo score is not positively correlated with embryonic development potential. Cytokines and proteins secreted by embryos are involved in the process of self-regulation and implantation of embryos, which may be used as markers to measure the developmental potential of embryos. This thesis will further study the expression of LH in human oocytes and preimplantation embryos on the basis of our previous work in order to provide experimental basis for exploring the regulatory role of LH in the development of human eggs and embryos. Since the 1950s, researchers have done a great deal of research on the evaluation of endometrial embryo acceptability from the aspects of histomorphology, subcellular structure, local factor expression and the regulation of steroid hormones.
