促红细胞生成素对大鼠肾缺血再灌注损伤的影响及机制

【摘要】:目的:本研究采用大鼠肾缺血再灌注损伤模型,探讨重组人促红细胞生成素(recombinant human erythropoietin, rHuEPO)对大鼠肾缺血再灌注损伤的作用,并对其可能机制进行初步研究。方法:雄性Wistar大鼠72只,随机分为三组:(1)假手术组(N=24),按取材时间不同又分为4个小组,即相当于再灌注1h、6h、12h、24h取材组,每组6只;(2)肾缺血再灌注损伤模型组(N=24),组内又分为再灌注1h、6h、12h、24h 4个时间点,每个时间点6只;(3) rHuEPO组(N=24),组内又分为再灌注1h、6h、12h、24h 4个时间点,每个时间点6只。将动物麻醉,开腹,假手术组切除右肾,不夹闭左肾动脉,模型组和rHuEPO处理组切除右肾,夹闭左肾动脉45min后松夹再灌,rHuEPO处理组在再灌注开始前尾静脉注射rHuEPO3000U/Kg,假手术组和模型组在再灌注开始前尾静脉注射等量生理盐水。再灌注达相应时间点时,从各小组大鼠下腔静脉取血3ml备测血尿素氮(BUN)和血肌酐(Cr),然后迅速切除左肾,称取0.5克肾组织迅速冷冻,待作成10%组织匀浆,测定丙二醛(MDA)、超氧化物歧化酶(SOD)和髓过氧化物酶(MPO),其余肾组织用10%甲醛固定,制备石蜡切片,作HE染色观察病理变化以及作免疫组化分析肾组织P-选择素和ICAM-1表达情况。结果:(1)肾功能改变:模型组除再灌注1h组血BUN无明显升高外,其余各小组BUN、Cr较假手术组明显升高(P0.01),rHuEPO处理组除再灌注1h组BUN与假手术组无明显差异外,其余各小组BUN较相应时间点假手术组明显升高(P0.05,P0.01),rHuEPO处理组中再灌注1h,12h,24h组血Cr较假手术组明显升高(P0.05,P0.01),而再灌注6h组血Cr与假手术组无显著差异。与肾IRI模型组比较,rHuEPO处理组再灌注6h、12h、24h组BUN、Cr明显降低(P0.01,P0.05),rHuEPO处理后再灌注1h组血BUN和Cr与模型组无明显差异。(2)肾组织生化指标检测结果:肾组织MDA、SOD测定结果:与假手术组比较,模型组各组肾组织MDA含量均明显升高(P0.05,P0.01),而各组肾组织SOD活力均显著降低(P0.05,P0.01),rHuEPO处理组除再灌注24h组MDA含量较假手术
[Abstract]:Aim: to investigate the effect of recombinant human erythropoietin (human erythropoietin, rHuEPO) on renal ischemia-reperfusion injury in rats and its possible mechanism. Methods: Seventy-two male Wistar rats were randomly divided into three groups. The sham operation group was divided into 4 groups according to the time of sampling. Each group (n = 6) was divided into 4 time points (1 h, 6 h, 12 h, 24 h) rHuEPO group (n = 6, n = 6) were divided into two groups (n = 6, n = 6). Animals were anesthetized, laparotomy, sham-operation group, right kidney was resected, left renal artery was not clipped, right kidney was resected in model group and rHuEPO treatment group. RHuEPO treatment group was injected with rHuEPO _ 3000U / kg before reperfusion, and the sham operation group and model group received the same amount of normal saline before reperfusion. Blood urea nitrogen bun (3ml) and creatinine (Cr) were collected from the inferior vena cava of the rats at the corresponding time point of reperfusion, and then the left kidney was quickly removed, and the kidney tissue was quickly frozen, and 10% tissue homogenate was prepared. The levels of malondialdehyde (MDA), superoxide dismutase (SOD) and myeloperoxidase (MPO) were measured. The other renal tissues were fixed with 10% formaldehyde to prepare paraffin sections. The pathological changes were observed by HE staining and the expression of P- selectin and ICAM-1 in renal tissues were analyzed by immunohistochemistry. Results (1) Renal function changes: except for 1 h reperfusion group, the blood BUN in the model group was significantly higher than that in the sham-operation group, except that the BUN in the model group was not significantly different from that in the sham-operation group, except that the BUN in the model group was significantly higher than that in the sham-operation group. The BUN of the other groups was significantly higher than that of the sham operation group at the corresponding time point. The serum Cr in the reperfusion group was significantly higher than that in the sham-operation group at 1h, 12h and 24h after reperfusion, but there was no significant difference between the six h reperfusion group and the sham-operation group. Compared with the renal IRI model group, the blood BUN and Cr in the rHuEPO treated group were significantly lower than those in the model group after reperfusion for 6 h and 12 h and 24 h after reperfusion. There was no significant difference in serum BUN and Cr between the model group and the model group. The results showed that: compared with the sham operation group, the blood BUN and Cr in the renal tissue of the RHuEPO treated group were significantly lower than those in the sham operation group, and the results were as follows: compared with the sham operation group, there was no significant difference in serum BUN and Cr between the model group and the model group. The content of MDA in renal tissue of model group was significantly higher than that of sham operation group, while the activity of SOD in renal tissue of each group was significantly lower than that of sham operation group except for 24 hours after reperfusion.
