碘缺乏和碘过量对大鼠心肌肌球蛋白重链基因mRNA表达的影响

【摘要】:碘是机体合成甲状腺激素(Thyroid Hormone,TH)所必需的微量元素,碘缺乏或碘过量都将影响TH的合成。TH有许多重要的生理功能,心脏是受TH调节的重要靶器官之一,而心肌肌球蛋白重链基因则是TH调节的重要靶基因。肌球蛋白是心肌主要的结构蛋白,也是起收缩作用的主要蛋白。心肌肌球蛋白重链(myosin heavy chain,MHC)包括α和β两条链。然而,碘缺乏和碘过量能否影响MHC基因的表达,,尚未见文献报道。 本实验通过低碘和高碘饲养,造成碘缺乏和碘过量动物模型,观察其对正常大鼠心肌α-MHC和β-MHC mRNA表达的影响。 材料与方法 实验动物:体重120-140g的健康Wistar大鼠90只。将动物随机分为6组:低碘组(LI),适量碘组(NI),5倍碘组(5HI),10倍碘组(10HI),50倍碘组(50HI),100倍碘组(100HI)。每组15只大鼠。 LI组动物饲以碘含量小于50μg/kg的低碘饲料,饮用去离子水。NI组和4个HI组大鼠饲以平均碘含量为300μg/kg的正常大鼠饲料;NI组饮用自来水,4个HI组饮用含不同高剂量碘酸钾(KIO_3)的自来水。计算6组大鼠日碘摄入量分别为:<1μg/d(LI组)、6.15μg/d(NI组)、30.75μg/d(5HI组)、61.5μg/d(10HI组)、307.5μg/d(50HI组)、615μg/d(100HI组)。 大鼠喂养6个月后,称重,抽血测定甲状腺激素,心电图检查,股动脉放血处死动物并迅速摘取心脏,液氮速冻后-80℃保存。 血清甲状腺激素的测定:TT_3、TT_4、FT_3、FT_4采用化学发光法。 心电图描记:LI组由于体格瘦小和麻醉易死未进行心电图检查。NI组和各

The effects of iodine deficiency and iodine excess on the expression of 伪 -MHC and 尾 -MHC mRNA in myocardium of normal rats were observed. Materials and methods Experimental animals: 90 healthy Wistar rats weighing 120-140 g. The animals were randomly divided into six groups: low iodine group (LI), moderate iodine group (n = 5) with 5 times iodine (n = 5) and 10 times iodine group (n = 10) with 50 times iodine (n = 50) and 100 times iodine group (n = 100). There were 15 rats in each group. The animals of Li group were fed with low iodine diet with iodine content less than 50 渭 g/kg. Rats of deionized water.
