林生地霉致病性的动物实验研究

【摘要】: 目的:Geotricum sivicola是地霉属新发现的一个种,目前国内外有关的报道极少。2002年我科从一位脓癣病人的病发及痂皮部位首次分离出此菌,由中国医学科学院微生物研究所鉴定为Geotricum sivicola,原翻译为昔维考拉地霉,现根据其拉丁文原意译为林生地霉。李秀丽等[1]从形态学、营养生理学、分子生物学等各个角度对林生地霉进行初步研究,并且通过皮肤感染的动物实验及毛发受侵试验对其致病力进行了初步探讨。 本研究通过不同途径给予不同免疫状态下的昆明小鼠注射林生地霉,检测是否能够引起系统性的林生地霉感染,并且观察不同免疫状态下动物的死亡情况,同时对自然死亡和分期处死的小鼠进行解剖,通过直接镜检、组织逆培养、组织病理学检查、组织载菌量测定等方法比较各脏器的感染程度以及电镜观察该菌在组织中的形态。为进一步了解此菌的致病力和引起的病理形态学改变提供实验室依据。 方法: 1林生地霉系统感染动物模型的制作将我科保存菌种复温,转种在沙堡氏培养基上37℃培养一周。用无菌生理盐水制备成菌悬液,通过皮下、腹腔、静脉三种途径接种至不同免疫状态的小鼠。 2死亡率的观察连续观察各组小鼠感染该菌后4周内的自然死亡情况,记录死亡日期和死亡只数。
[Abstract]:Objective: Geotricum sivicola is a newly discovered species of Trichoderma, which is rarely reported at home and abroad. In 2002, the strain was first isolated from the hair and callus of a patient with psoriasis purpura. Geotricum sivicola was identified by the Institute of Microbiology, Chinese Academy of Medical Sciences. Li Xiuli et al. [1] the pathogenicity of Trichoderma woodland was studied from the aspects of morphology, nutrition physiology and molecular biology, and its pathogenicity was studied by animal experiment of skin infection and hair invasion test. In this study, Kunming mice in different immune states were injected with Trichoderma plantarum in different ways to detect whether they could cause systemic infection of Trichoderma plantarum, and to observe the death of animals in different immune states. At the same time, the mice died by stages were dissected. The infection degree of the organs and the morphology of the bacteria in the tissues were observed by direct microscope examination, tissue reverse culture, histopathological examination and tissue carrying capacity measurement. To provide laboratory basis for further understanding the pathogenicity and pathomorphological changes caused by this bacterium.
