前列腺增生大鼠模型的建立
[Abstract]:Aim: to investigate the effect of testosterone propionate on prostatic hyperplasia in ovariectomized and non-castrated SD rats. Methods Twenty-five male Sprague-Dawley rats aged 8 weeks were randomly divided into normal control group (n = 5), no castration model group (n = 15) and ovariectomized model group (n = 5), and no castration model group was divided into no castration low dose group, middle dose group and high dose group according to the dose of testosterone propionate injection. The rats in ovariectomized group were injected with testosterone propionate 5 mg/ (kg d); for 1 week, and testosterone propionate was injected continuously for 4 weeks in the ovariectomized model group and the non-castrated model group. The prostatic gland was separated after the last administration and its wet weight was measured. Volume and organ coefficient, HE staining of tissue sections, changes of prostate tissue under light microscope. Results: the wet weight, volume and organ coefficient of prostate were increased in the low dose group and middle dose group. Compared with the normal control group, the prostate volume and organ coefficient of the non-ovariectomized high dose group and ovariectomized group were significantly higher than that of the normal control group (P0.05). The difference was statistically significant (P0.01), the volume of prostate and the size of glandular cavity in ovariectomized group and non-castrated group were larger than those in normal control group, and the boundary between prostate gland and surrounding tissue in non-castrated group was more clear than that in non-castrated group. The intraglandular papillary process was also more pronounced than that in the castrated group. Conclusion: subcutaneous injection of testosterone propionate for 5 mg/ (kg d) can effectively establish a rat model of benign prostatic hyperplasia (BPH).
