家兔下肢缺血模型的制作

【摘要】: 目的:模拟人的动脉硬化闭塞症,制作一种下肢缺血的实验动物模型,建立持久的、稳定的缺血模型。 方法:选取雄性新西兰白兔10只,结扎并切除左下肢从腹股沟韧带至膝关节的股动脉及其分支,造成左下肢缺血,术后按时对家兔患肢缺血程度及肢体变化予以评估,直至术后6周。 结果:家兔缺血侧肢体一周以后开始出现肌肉萎缩,坏死,肢体溃疡,,指甲增长、增厚等。术后家兔患肢血压明显下降,左下肢/右下肢血压率显著下降(P<0.05)。放射性同位素灌注扫描示:患肢血流量明显减少,左下肢/右下肢血流量明显降低,左下肢同位素兴趣图分布明显低于右下肢。DSA血管造影示:术后左下肢/右下肢的细微侧枝血管记数率从1.00±0.52增加至3.00±0.04,血管数量明显增加(P<0.05)。术后6周患侧与健侧肌肉的病理学观察示:患侧肌纤维变小、变少、血管侧支增多。 结论:家兔下肢慢性缺血模型成功制作,与临床动脉硬化闭塞症病例症状相似,表现为持久的、稳定的左下肢缺血。
[Abstract]:Aim: to establish an experimental animal model of lower extremity ischemia by simulating human arteriosclerosis obliterans. Methods: ten male New Zealand white rabbits were selected and the femoral artery and its branches from inguinal ligament to knee joint were ligated and resected to cause ischemia of the left lower extremity. Up to 6 weeks after operation. Results: muscle atrophy, necrosis, ulcers, nail growth and thickening began to occur in the ischemic limbs after one week. After operation, the blood pressure of the affected limbs decreased significantly, and the blood pressure rate of the left / right lower limbs decreased significantly (P < 0.05). Radioisotope perfusion scan showed that the blood flow of the affected limb was significantly decreased, and that of the left / right lower extremities was significantly decreased. The distribution of isotopic interest map of left lower extremity was significantly lower than that of right lower extremity. DSA showed that the count rate of fine lateral branches of left lower limb / right lower extremity increased from 1.00 卤0.52 to 3.00 卤0.04, and the number of blood vessels increased significantly (P < 0.05). The pathological observation of the affected and healthy muscles at 6 weeks after operation showed that the muscle fibers of the affected side became smaller and less, and the collateral branches of the vessels increased. Conclusion: the model of chronic ischemia of rabbit's lower extremities is made successfully, which is similar to the clinical symptoms of arteriosclerosis obliterans, showing persistent and stable left lower extremity ischemia.
