人B淋巴细胞刺激因子载体稳定性及其嵌合毒素的表达载体构建与产物性质鉴定

【摘要】:人B淋巴细胞刺激因子(human B Lymphocyte Stimulator,hBlyS)是一种B淋巴细胞的共刺激因子,属TNF超家族成员。BLyS有三个受体BAFF-R,BCMA和TACI,三个受体主要表达于B淋巴细胞。BLyS在Anti-IgM或IL-4存在下能专一的刺激B细胞增殖、分化及分泌免疫球蛋白,在体液免疫中有重要的作用。BLyS同自身免疫性疾病有关,在很多的淋巴瘤的发生中有作用。 嵌合毒素是具有导向能力的分子(载体)和具有细胞毒性的分子(毒素)偶联而成的具有特异性细胞杀伤能力的杂合分子。载体主要使用抗体及细胞因子,毒素主要使用超抗原及蛋白质合成抑制剂等。嵌合毒素在肿瘤的治疗上有其突出的优点:对肿瘤细胞具有选择性杀伤作用,细胞毒效价高,无抗药性。 本论文详细介绍:本室构建的重组pET30a/hsBLyS质粒在BL21(DE3)菌株中的稳定性观察,嵌合毒素hsBLyS-PE40的基因构建、表达及活性检测。
[Abstract]:Human B lymphocyte stimulating factor (human B Lymphocyte StimulatorhBlyS) is a costimulator of B lymphocytes. BLyS, a member of the TNF superfamily, has three receptors, BAFF-RCMA and TACI.The three receptors are mainly expressed in B lymphocytes. BLyS can specifically stimulate the proliferation, differentiation and secretion of immunoglobulin in B cells in the presence of Anti-IgM or IL-4. BLyS plays an important role in humoral immunity. BLyS is associated with autoimmune diseases and plays an important role in the development of lymphoma. Chimeric toxin is a hybrid molecule with specific cytotoxicity, which is conjugated with molecules (vectors) and cytotoxic molecules (toxins). Antibodies and cytokines are used as carriers, superantigens and protein synthesis inhibitors are used as toxins. Chimeric toxin has many advantages in the treatment of tumor: selective killing of tumor cells, high cytotoxicity and no drug resistance. The stability of recombinant pET30a/hsBLyS plasmid in BL21 (DE3) strain, gene construction, expression and activity detection of chimeric toxin hsBLyS-PE40 were described in detail.
